摘要
目的探讨选择性PDGFR抑制剂CP-673451对人非小细胞肺癌(NSCLC)细胞增殖、侵袭、迁移的影响及其作用机制。方法体外培养人非小细胞肺癌A549细胞,采用Transwell法、划痕法和免疫荧光法检测细胞侵袭、迁移和片状伪足形成的影响,Western印迹实验检测A549细胞的Akt、GSK-3β、p70S6和S6蛋白的磷酸化水平。结果CP-673451能够抑制A549细胞侵袭、迁移和伪足形成,抑制Akt、GSK-3β、p70S6和S6蛋白的磷酸化,且呈浓度依赖性。结论CP-673451抑制人非小细胞肺癌细胞侵袭和迁移,抑制PDGFR下游信号传导通路蛋白的表达,可能是其作用机制。
Objective To investigate the effects of selective PDGFR inhibitor CP-673451 on proliferation,invasion and migration of human non-small cell lung cancer(NSCLC)cells and its mechanism.Methods Human non-small cell lung cancer A549 cells were cultured in vitro.Cell invasion,migration and sheet pseudopod formation were detected by using the Transvel methode,the scratch and the immunofluorescence experiments.The phosphorylation levels of Akt,GSK-3β,p70S6 and S6 proteins were detected by using Western blotting.Results CP-673451 inhibited A549 cells of invasion,migration and pseudopod formation,and the phosphorylated of Akt,GSK-3β,p70S6 and S6 in a concentration-dependent manner.Conclusion CP-673451 inhibits the invasion and migration of human non-small cell lung cancer cells,and may suppress the expression of downstream signaling transduction pathway proteins of PDGFR,which may be the potential mechanism.
