摘要
近年来,屡见高致病性H5N6禽流感病毒由禽跨种感染人及其他哺乳动物类的报道。而临床用药的盲目性导致其预后不良,本试验以评价流感病毒NA抑制剂奥司他韦是否能够有效抑制该病毒为试验目的。通过构建H5N6小鼠感染模型、制定奥司他韦治疗方式及药物用量以评价奥司他韦对H5N6流行株的抑制作用。通过存活率分析,预防+治疗组奥司他韦对H5N6有较好预防效果,而单纯治疗组小鼠存活率只达50%。通过肺组织中病毒含量结果显示在攻毒第12日未能检测到病毒,而治疗组滴度下降至2.500×log10 EID50/0.1mL(*P<0.01)。由此可见,奥司他韦对H5N6亚型禽流感小鼠适应株有较好的预防效果,对于攻毒后再治疗可一定程度抑制病毒的复制。
In recent years, highly pathogenic H5N6 avian influenza virus has been reported to cross-species from birds to infect humans and other mammals. The blindness of clinical medication leads to poor prognosis. This experiment aims to evaluate whether oseltamivir, an influenza virus NA inhibitor, can effectively inhibit the virus. The inhibiting effect of oseltamivir on H5N6 epidemic strain was evaluated by establishing the infection model of H5N6 mice, formulating the treatment method and dosage of oseltamivir. The results showed that the prevention treatment group had better preventive effect on H5N6 through survival analysis, while the survival rate of mice in the treatment group alone was only 50%. The results of virus titer in lung tissue in the prevention group dropped below the detection line on the 12th day of challenge, while that in the treatment group decreased to 2.500×log10 EID50/0.1 mL(*P<0.01). Therefore, oseltamivir has a good preventive effect on H5N6 avian influenza virus in mice and can inhibit the replication ability of the virus to a certain extent after treatment.
