摘要
肺动脉高压(PH)是一种由异源性疾病和不同发病机制引起的以肺动脉压力增高为主要表现的疾病状态,严重者出现右心衰竭,预后差。Gremlin是一种细胞外糖蛋白,作为骨形态形成蛋白(BMP)的拮抗剂,在胚胎气道和肺循环发育中发挥了关键作用。本文综合分析Gremlin在PH发生发展中的作用,发现Gremlin通过抑制肺动脉内皮细胞损伤修复、促进平滑肌细胞增殖、迁移,诱导细胞外基质沉积及炎症细胞浸润,参与PH的发生发展过程,成为PH治疗的新的靶点。
Pulmonary hypertension (PH) is characterized by sustained increase of pulmonary arterial pressure and eventually leading to right heart failure.PH is caused by heterogeneous diseases and different pathogenesis.Gremlin is an extracellular glycoprotein that was first identified over a decade ago through its important role in embryonic development, in which it acts as an antagonist of bone morphogenetic protein actions.It plays a critical role in the development of normal airways and the pulmonary circulation in the embryo.The article reviews that Gremlin contributes to the pathogenesis of pulmonary hypertension by inhibiting the repair of pulmonary artery endothelial cells, promoting smooth muscle cell proliferation and migration, inducing extracellular matrix deposition and infiltration of inflammatory cells which suggests future avenues of research.
