金刚藤颗粒对急性盆腔炎模型大鼠NLRP3炎症小体通路及免疫功能的影响 被引量:6

摘要 目的观察金刚藤颗粒对急性盆腔炎(APID)模型大鼠NLRP3炎症小体通路及免疫功能的影响。方法将雌性SD大鼠105只随机分为假手术组、模型组及金刚藤颗粒低、中、高剂量组,每组均15只,灌胃治疗15 d后,ELISA检测大鼠血清白细胞介素-1β(IL-1β)、白细胞介素-18(IL-18)水平;苏木精-伊红(HE)染色观察子宫组织的病理形态;实时定量PCR及Western blotting分别检测NLRP3、ASC、IL-1β、Caspase-1 mRNA及蛋白表达水平。结果 HE染色显示,模型组大鼠子宫内膜上皮细胞脱落坏死,腔壁结构紊乱,全层炎性细胞浸润,有充血水肿;与模型组比较,金刚藤颗粒治疗组症状较模型组有显著改善;与对照组、假手术组比较,模型组血清IL-1β、IL-18水平显著升高(P <0.05),与模型组比较,金刚藤颗粒治疗组IL-1β、IL-18水平显著降低(P <0.05);与对照组比较,模型组NLRP3、ASC、IL-1β、Caspase-1 mRNA水平显著升高(P <0.05),与模型组比较,金刚藤颗粒治疗组NLRP3、ASC、IL-1β、Caspase-1 mRNA水平显著降低(P <0.05);模型组NLRP3、ASC、IL-1β、Caspase-1蛋白表达水平显著高于假手术组(P <0.05),金刚藤颗粒治疗组NLRP3、ASC、IL-1β、Cas-pase-1蛋白表达水平较模型组显著降低(P <0.05)。结论金刚藤颗粒对急性盆腔炎模型大鼠具有显著治疗作用,其可能通过抑制NLRP3炎症小体通路发挥免疫作用。 Objective:To investigate the effects of Jingangteng Granule on NLRP3 inflammatory body pathway and immunologic function of rats with acute pelvic inflammatory disease(APID). Methods:105 healthy female SD rats were randomly divided into the sham operation group,the model group and low,medium,and high dose Jingangteng Granule groups. They were given intragastric administration for 15 d. The levels of serum IL-1β and IL-18 were detected by ELISA;the pathological morphology of uterus was observed by HE staining;the expression levels of NLRP3,ASC,IL-1β and Caspase-1 were detected by real-time quantitative PCR and Western blotting.Results:HE staining showed that the endometrial epithelial cells of the model group were exfoliated and necrotic;the wall structure was disordered;the whole layer of inflammatory cells infiltrated,and there were congestion and edema. Compared with the model group,the symptoms of Jingangteng Granule treatment groups were significantly improved. The levels of serum IL-1β and IL-18 in the model group were significantly higher than those in the control group and the sham operation group(P < 0.05);compared with the model group,those in Jingangteng Granule treatment group were significantly lower(P < 0.05). Compared with the control group,the levels of NLRP3,ASC,IL-1β and Caspase-1 in the model group increased significantly(P < 0.05);compared with the model group,those in Jingangteng Granule treatment groups were significantly decreased(P < 0.05);those in the model group were significantly higher than those in the sham-operated group(P < 0.05),and the levels of each index in the Jingangteng Granule treatment group were significantly lower than those in the model group(P < 0.05). Conclusion:Jingangteng Granule has a significant therapeutic effect on acute pelvic inflammation model rats,which may play an immune role by inhibiting NLRP3 inflammatory body pathway.
