膀胱穿刺活检术在膀胱出口部分梗阻大鼠中应用的可行性

摘要 目的本实验创新性地将膀胱穿刺活检技术应用于膀胱出口部分梗阻大鼠以评价其可应用性。方法雌性SpragueDawley大鼠18只行尿道外口结扎法造模后,根据不同梗阻时间分为对照组、梗阻14d组、梗阻28d组,利用穿刺技术提取膀胱组织标本,行HE染色观察膀胱形态学变化,通过免疫组织化学染色观察机械敏感K+通道(TERK-1)的表达趋势,判断穿刺方式获取标本的实际应用价值。结果从膀胱穿刺所取得的标本可清楚观察到膀胱出口梗阻后膀胱的典型变化,HE染色下观察到梗阻14d组、梗阻28d组大鼠的膀胱肌纤维束排列及形态被破坏,肌细胞肥大,平滑肌横截面明显增厚。免疫组化提示,随梗阻时间延长,TERK-1通道表达量呈下降趋势,差异具有统计学意义(P=0.035)。结论膀胱穿刺术以微创的方法成功收集到足够的组织标本,用于形态学及病理学检测,是一种可行的方法。 Objective In this study,we innovatively applied bladder needle biopsy technique to rats with partial bladder outlet obstruction(BOO)in order to evaluate its applicability.Methods BOO models were established in18 female SpragueDawley rats which were divided into control group,14-day BOO group and 28-day BOO group.After the bladder tissues were obtained with needle biopsy technique,HE staining was conducted,and the morphological changes of bladder tissues were observed.The expression of TWIK related K+channel 1(TERK-1)was determined.Results We could clearly observe the typical morphological changes after the bladder outlet was partially obstructed.HE staining showed that the arrangement and morphology of bladder smooth muscle fiber bundles were destroyed in the 14-d BOO group and28-d BOO group,the muscle cells hypertrophied,and the cross-section of smooth muscle thickened.Immunohistochemistry indicated that the expression of TERK-1 decreased with the time of obstruction,and the difference was statistically significant(P=0.035).Conclusion Bladder needle biopsy technique is able to collect enough bladder tissue specimens for pathomorphological analysis.
