摘要
目的探索诱导多能干细胞来源心肌细胞(iPS-CMs)移植对心肌梗死后小鼠心功能的作用。方法以无饲养层培养法培养iPS细胞并诱导分化成iPS-CMs。肌钙蛋白T(cTnT)和肌球蛋白重链(MHC)免疫荧光鉴定iPS-CMs。30只C57 BL/6小鼠随机分为Sham组、PBS组和iPS-CMs组。结扎小鼠冠状动脉前降支制作心肌梗死模型,分别在心梗及心梗边缘区注射总量10μL 4×10~5 iPS-CMs(iPS-CMs组)和等量的PBS(PBS组),细胞移植后4周行心脏超声检测心功能。结果 cTnT和MHC免疫荧光结果显示成功培养分化出iPS-CMs。心脏超声结果显示移植4周后,与PBS组相比,iPS-CMs组小鼠心功能改善明显。结论 iPS-CMs移植能改善心肌梗死小鼠的心功能。
Objective To investigate the repair effect of translation of induced pluripotent stem cells-derived cardiomyocytes for cardiac function of mouse of myocardial infarction.Methods iPS cells(Induced pluripotent stem cells)were cultured in a feeder-independent manner,and iPS cells was induced to differentiate into cardiomyocytes using EB(Embryo body)method.Immunofluorescence stained for cTnT and MHC was used to identify the iPS-CMs.A number of 30 mouse were randomly divided into sham group,PBS group and iPS-CMs group.Myocardial infarction mouse model was established by ligating the left anterior descending coronary artery,Sham operation was performed by passing the needle through the heart tissue,but the suture was not ligated.10 μL of PBS or iPS cells were transplanted into two adjacent positions of the ischemic area intramyocardially through a 30-gauge needle.4 weeks after the operation,cardiac function was detected by usig transthoracic echocardiography.Results cTnT and MHC positive cells can be seen in differentiated cells.At 4 weeks after myocardial infarction and treatment of cell transplantation,transthoracic echocardiography results showed that mouse of iPS-CMs group promoted cardiac contractile function recovery of infarcted heart better than PBS group.Conclusion Translation of induced pluripotent stem cells-derived cardiomyocytes improve cardiac function of mouse of myocardial infarction.
