摘要
目的:探讨参芪复方对糖尿病GK大鼠肠道菌群及血糖波动的影响。方法:建立由高脂饲料喂养的糖尿病大血管病变大鼠模型。实验分4组,Wistar组,模型组,参芪复方组,西药组。于灌胃后每周1次测1日内8∶00,10∶00,14∶00,16∶00,18∶00五次血糖水平,并用血糖稳定性指标(MBG、SDBG、LAGE)量化各组血糖稳定性;检测血清GLP-1水平;运用16SrDNA基因测序分析各组大鼠肠道菌群。结果:参芪复方组可减少大鼠血糖波动,升高血清GLP-1水平,增加拟杆菌门/厚壁菌门比值,升高拟杆菌属、丁酸菌属、Blautia菌属、罗氏菌属的相对丰度。结论:具有养阴益气活血功效的参芪复方可调节肠道菌群,增加产丁酸菌的相对丰度,促进GLP-1分泌,达到调节血糖稳态,防治糖尿病大血管病变的作用。
Objective: To explore the mechanism of Shenqi Compound on intestinal microflora and blood glucose fluctuations in diabetic GK rats. Methods: We established a rat model of diabetic macroangiopathy fed with high-fat-diet. The model rats were randomly divided into Wistar group,model group,Shenqi group,Sitagliptin group( referred to as the Western medicine group),a total of 4 groups. Starting from the gavage administration once a week measured at 8:00,10:00,14:00,16:00,18:00 five-point blood glucose levels. The blood glucose stability was quantified with the indexes of MBG,SDBG and LAGE. We detected GLP-1 level in serum. The intestinal flora of rats was analyzed by16 SrDNA gene sequencing. Results: Shenqi Compound can reduce blood glucose fluctuations in rats,increase serum GLP-1 levels,increase Bacteroides/Firmicutes ratio,raise Bacteroides,Butyricimonas,Blautia and Roseburia relative abundance. Conclusion: Shenqi Compound Recipe which has the function of supplenmenting Qi and Yin and promoting the blood circulation,may achieve the function of regulating blood glucose homeostasis and preventing and treating diabetic macroangiopathy by regulating the intestinal microflora,increaseing the relative abundance of butyrate-producing bacteria,promoting the secretion of GLP-1.
