摘要
为了研制经济环保的新型鸡球虫卵囊漂浮剂,试验配制了不同浓度的蔗糖溶液,对柔嫩艾美耳球虫卵囊进行漂浮试验。结果显示:0.9~1.2mol/L的蔗糖溶液对球虫卵囊的漂浮效果较好,漂浮率达92%以上,卵囊沉淀时应将蔗糖溶液稀释至0.3mol/L以下。试验还选择了几种实验室常见试剂,配制成密度在1.15~1.20g/mL之间的溶液进行柔嫩艾美耳球虫卵囊漂浮回收及孢子化试验,结果显示40%磷酸氢二钾溶液可以替代饱和食盐水和蔗糖溶液用于鸡球虫卵囊的漂浮纯化。
In order to develop a new economically and environmentally friendly flotation agent for chicken coccidial oocysts,sucrose solutions of different concentrations were prepared and the flotation tests of eimeria tenella oocysts were carried out.The results showed that the 0.9-1.2 mol/L sucrose solution had a better effect on the flotation of coccidial oocysts,with a floating rate of more than 92%.Sucrose solution should be diluted to less than 0.3 mol/L when oocyst is deposited.Several common laboratory reagents were selected to prepare a solution with a density of 1.15-1.20 g/mL for floating recovery and sporulation of eimeria tenella oocysts.The results showed that 40%dipotassium hydrogen phosphate solution could replace saturated salt water and sucrose solution for floating purification of chicken coccidia oocysts.
