HIV-1感染过程中外周血CD8^+CD127^+T细胞活化耗竭研究 被引量:2

摘要 目的:检测HIV-1感染者CD8^+CD127^+T细胞CD38、HLA-DR和PD-1的表达水平,探讨HIV-1感染过程中CD8^+CD127^+T细胞免疫活化和耗竭的意义。方法:采集未经抗病毒治疗的HIV-1感染者63例及健康对照者16例,采用流式细胞术检测外周血单个核细胞中CD8^+CD127^+T细胞CD38、HLA-DR和PD-1表达水平。结果:与健康对照组比较,HIV-1典型进展者和慢性感染者CD8^+T细胞CD127的表达显著下降,CD8^+CD127^+T细胞CD38、HLA-DR、PD-1的表达显著升高( P <0.05);HIV-1感染者CD4计数与CD127、CD38、HLA-DR、PD-1的表达频率负相关;HIV-1感染者CD8^+CD127^+T细胞HLA-DR的表达频率与PD-1的表达正相关。结论: HIV-1感染过程中CD8^+CD127^+T细胞高活化耗竭是引起CD8^+CD127^+T细胞减少的原因。 Objective: To identify the status of CD8^+CD127^+ T cells during HIV-1 infection by detecting the expression levels of CD38, HLA-DR and PD-1 in CD8^+CD127^+ T cells. Methods: Peripheral blood mononuclear cells were isolated by density gradient centrifugation from 63 patients and 19 healthy donors.The CD8^+CD127^+T cell counts and CD38,HLA-DR,PD-1 expressions on CD8^+CD127^+T cells were analysed by flow cytometry. Results: Compared with healthy donors,HIV-1 infection resulted in a decrease of the CD127 on CD8 + T cells proportion in typical progressors and chronic persons.The expression of CD38,HAAL-DR,PD-1 on CD8^+CD127^+T cells in HIV-1 patients was significantly higher than that in healthy donors;the expression of CD127,CD38,HLA-DR,PD-1 negatively correlated with CD4^+T cell counts;PD-1 was positively correlated with HLA-DR expression on CD8^+CD127^+T cells in HIV-1 infected patients. Conclusion: Disease of CD8^+CD127^+T cells was in parallel with high level of immune activation and exhaustion.
