摘要
目的研究乳腺癌细胞中microRNA(miRNA)的表达与乳腺癌阿霉素化疗耐药性产生的相关性。方法选择人乳腺癌细胞株(MCF7)及人乳腺癌阿霉素耐药细胞株(MCF7/ADM)进行培养,提取其中的微小核糖核酸(microRNA)分子,运用RealTimeQ-PCR技术定量分析miR-100的表达水平,比较两种细胞中的miR-100表达情况。结果MCF7细胞株中的miR-100表达量低于MCF7/ADM细胞株中的miR-100表达量,差异有统计学意义(P<0.05)。结论miR-100的异常表达与乳腺癌化疗耐药性的发生存在正相关,本研究发现可以为乳腺癌的早期诊治与预后的生物标志物研究提供数据依据。
Objective To study the correlation between the expression of microRNA(microRNA) in breast cancer cells and the drug resistance to adriamycin in breast cancer. Methods Human breast cancer cell line(MCF7) and human breast cancer doxorubicin resistant cell line(MCF7/ADM) were cultured to extract microRNA molecules. Real Time Q-PCR was used to quantitatively analyze the expression level of microRNA-100 and compare the expression of microRNA-100 in the two cells. Results The expression of microRNA-100 in MCF7 cell line was lower than that in MCF7/ADM cell line, and the difference was statistically significant(P<0.05). Conclusion Abnormal expression of microRNA-100 is positively correlated with the occurrence of chemotherapy resistance in breast cancer. The findings of this study can provide data for the early diagnosis, treatment and prognosis of breast cancer.
