摘要
四氢生物蝶吟是芳香族氨基酸発化酶的必需辅酶,其合成不足或代谢缺陷会导致动物罹患多种神经性疾病,6-丙酮酰四氢蝶吟合酶是四氢生物蝶吟合成代谢途径中的关键酶。本研究采用生物信息学方法,对意大利蜜蜂6-丙酮酰四氢蝶吟合酶编码蛋白的氨基酸组成、理化性质、疏水性、跨膜区、结构功能域以及空间二级结构和三级结构等方面进行分析和预测,并构建了系统进化树。分析结果显示,意蜂6-丙酮酰四氢蝶吟合酶基因全长是829bp,含有一个429bp的开放阅读框,编码142个氨基酸,属于亲水性蛋白,即非分泌蛋白,二级结构主要由a-螺旋构成,系统进化树分析结果与传统的生物学分类结果相符。实验结果可为进一步开展意蜂6-丙酮酰四氢蝶吟合酶基因家族的功能研究和应用提供信息参考。
In this study, we analyzed and predicted the composition of amino acid sequence, physical and chemical properties, hydrophobicity, transmembrane domain, structural domain, secondary structure and tertiary structure of the Apis mellifera 6-pyruvoyl tetrahydrobiopterin synthase (PTS) by bioinformatics methods, and also constructed the phylogenetic tree. The results showed that 6-pyruvoyl tetrahydrobiopterin synthase (PTS) from Apis mellifera had the total length of 829bp, encoded 142 amino acid, belonged to hydrophilic protein, and its secondary structure was mainly composed of a-helix. The results of phylogenetic tree analysis were consistent with the results of traditional biological classification. These experiment results would provide information references for further function research and application of Apis mellifera 6-pyruvoyl tetrahydrobiopterin synthase gene family.
