摘要
目的:探究不同产地和生长年限甘草中甘草苷和甘草酸的含量,为有效控制其质量提供依据。方法:以甘草中有效成分甘草苷和甘草酸为检测指标,采用HPLC法比较不同产地和生长年限甘草中这两种成分的含量,从化学成分角度来探讨中药的质量。色谱条件:色谱柱:Kromasil C18色谱柱(4.6 mm×250 mm,5μm);流速:1.0 mL/min;检测波长:甘草苷276 nm,甘草酸254 nm;柱温:28℃;流动相:乙腈-0.05%磷酸水梯度洗脱系统。结果:野生甘草质量最好;白塔三年生甘草次之;内蒙古3年生甘草较好;岷县2年生甘草质量较差,瓜州甘草质量最差。结论:根据上述测定结果,采用HPLC方法测定甘草中甘草苷和甘草酸的含量,操作简单,可靠,重现性好,可作为甘肃道地药材甘草质量控制的有效方法。
Objective:To determinate of glycyrrhizin and glycyrrhizic acid of Radix Glycyrrhizae at different origin and growth,which could evaluate the quality of Radix Glycyrrhizae,and provided a reliable method. Methods:The main active ingredient glycyrrhizin and glycyrrhizin of Radix Glycyrrhizae were detected by HPLC method to compare the content of two components of Radix Glycyrrhizae with different origins and growth years.The chromatographic conditions were used:Column:Kromasil C18 column(4.6 × 250 mm,5 μm),flow rate:1.0 mL/min,detection wavelength:glycyrrhizin 276 nm,glycyrrhizic acid 254 nm,column temperature:28 ℃,mobile phase:acetonitrile-0.05% phosphoric acid gradient elution system. Results:The quality of wild Radix Glycyrrhizae was the best,triennial Radix Glycyrrhizae from White tower and triennial Radix Glycyrrhizae from Inner Mongolia were better. The quality of biennial Radix Glycyrrhizae from Min county was poor,that from Guazhou was the worst.Conclusion:HPLC detection method was simple,accurate,good reproducibility and high precision,which could be used as the quality control method of Radix Glycyrrhizae.
