Expression and Clinical Significance of ALDH1 and Nestin in Ovarian Cancer

目的研究ALDH1和Nestin在卵巢癌中的表达及临床意义。方法应用组织芯片及免疫组化法,检测130例卵巢癌组织及同期30例正常卵巢组织中ALDH1和Nestin的表达情况,并结合卵巢癌不同临床病理参数和患者预后进行比较分析。结果 ALDH1和Nestin在卵巢癌组织中的阳性表达率均显著高于正常卵巢组织(均P<0.05)。ALDH1阳性表达与卵巢癌的组织学类型、分级、淋巴结转移、FIGO分期及Ki-67指数显著相关(均P<0.05);Nestin阳性表达与卵巢癌的分级、淋巴结转移及FIGO分期显著相关(均P<0.05)。相关性分析显示ALDH1的表达与Nestin的表达呈正相关(P<0.05)。Kaplan-Meier生存曲线显示ALDH1阳性表达的卵巢癌患者生存率显著低于阴性表达组(P<0.05),ALDH1和Nestin双阳性表达的卵巢癌患者生存率显著低于ALDH1和Nestin单阳性表达或双阴性表达的卵巢癌患者(P<0.05)。结论 ALDH1和Nestin在卵巢癌组织中高表达,可能促进了卵巢癌的发生发展,两者联合检测为卵巢癌早期诊断、治疗和预后判断提供参考依据。

Objective To explore the expression of ALDH1 and Nestin in ovarian cancer and their correlations with the prognosis.Methods The expression levels of ALDH1 and Nestin in 130 cases of ovarian cancer and 30 cases of normal ovarian tissues during the same period were detected by tissue microarray and immunohistochemical method.And correlation between different clinical pathological parameters of ovarian cancer and patient prognosis was analyzed.Results The positive expression rates of ALDH1 and Nestin in ovarian cancer tissues were significantly higher than those in normal ovarian tissues(both P<0.05).ALDH1 positive expression was significantly correlated with the histological type,tumor grade,lymph node metastasis,FIGO stage and Ki-67 index of ovarian cancer(all P<0.05).Nestin positive expression was significantly correlated with the grade of ovarian cancer,lymph node metastasis and FIGO stage(all P<0.05).Correlation analysis showed that the expression of ALDH1 was positively correlated with the expression of Nestin(P<0.05).The Kaplan-Meier survival curve showed that the survival rate of ALDH1-positive ovarian cancer patients was significantly lower than that of the negative expression group(P<0.05).The survival rate of ovarian cancer patients with double positive expression of ALDH1 and Nestin was significantly lower than that of ovarian cancer patients with single positive or double negative expression of ALDH1 and Nestin(P<0.05).Conclusion The high expression of ALDH1 and Nestin in ovarian cancer may promote the development of ovarian cancer.The combined detection of ALDH1 and Nestin may be a molecular marker for ovarian cancer tissues and can be used to judge prognosis.
