摘要
为了提高水牛的繁殖效率,探明骨形态发生蛋白15(BMP15)在水牛生殖中的调控机制,试验以3,4,5,6月龄水牛睾丸组织为研究对象,采用免疫组化染色法分析BMP15及其受体激活素受体样激酶5(ALK5)的表达定位。结果表明:在3月龄组中BMP15免疫阳性产物主要定位于精细管上皮的精原细胞中;而4月龄组在精原细胞和间质细胞中均有表达定位;5月龄组主要定位于圆形精子细胞和变形精子中,睾丸间质细胞和精原细胞中均未检测到免疫阳性产物的存在;6月龄组又重新在精细管上皮的精原细胞和圆形精子细胞中检测到了BMP15免疫阳性产物。ALK5免疫阳性产物也定位于精原细胞、精子细胞和间质细胞中。说明BMP15可能参与精子的发生周期,在精子发生和成熟过程中起到一定作用。
In order to improve the reproductive efficiency of buffalo and to explore the regulation mechanism of bone morphogenetic protein 15 ( BMP15) in the reproduction of buffalo,testis tissue of 3,4,5,6 month-old buffalo was used as research object in this experiment and immunohistochemistry method was adopted for the analysis of the expression and localization of BMP15 and its receptor protein ALK5. The results showed that in 3 month-old group BMP15 was mainly located in spermatogonial cells of the epithelium of seminiferous tubules;in 4 month-old group it was expressed in spermatogonial cells and interstitial cells;in 5 month-old group it was mainly located in round spermatid and deformed sperm and no positive products were detected in testis interstitial cells and spermatogonial cells;in 6 month-old group BMP15 positive products were detected again in spermatogonial cells of the epithelium of seminiferous tubules and round spermatid. ALK5 positive products were also located in spermatogonial cells,spermatid and interstitial cells. The results suggested that BMP15 may be involved in spermatogenesis cycle and play a role in spermatogenesis and maturation.
