摘要
目的:探究以传统中医古方-"柴胡疏肝散"为基础,通过现代工艺得到的柴胡、白芍、枳壳、香附、延胡索、甘草6味中药有效部位提取物配伍组方改善大鼠肝郁气滞型功能性消化不良(functional dyspepsia,FD)的机制。方法:Wistar大鼠96只,雌雄各半,随机分为空白组、模型组、吗丁啉27 mg/kg组、柴胡疏肝散有效部位189、378、756 mg/kg组,采用慢性夹尾应激结合饮食失节的方法建立FD大鼠模型,采用定量方法观察测定FD大鼠胃排空率、小肠推进率、胃残留率。采用酶联免疫分析方法(ELISA)测定血清中胃促生长素(Ghrelin)、胃动素(MTL)及血管活性肠肽(VIP)。并应用蛋白印迹法检测大鼠胃组织紧密连接蛋白(ZO-2)、Yes-associated protein (YAP)、降钙素基因相关肽(CGRP)蛋白表达。结果:与空白组比较,模型组大鼠胃排空率和小肠推进率、Ghrelin、MTL水平明显降低,VIP含量明显升高,ZO-2蛋白明显下调,YAP、CGRP蛋白明显上调。与模型组比较,柴胡疏肝散有效部位各剂量组胃排空率、小肠推进率明显提高,Ghrelin、MTL分泌明显增加,而血清VIP含量明显降低,ZO-2蛋白水平显著上调,YAP、GCRP水平则显著下调。结论:柴胡疏肝散有效部位组方能够有效改善大鼠肝郁气滞型功能性消化不良,调节肠胃激素分泌以及相关蛋白表达。
Objective: This study aimed to investigate the effects of combinations of Chaihu, Baishao, Zhike, Xiangfu,Yanhusuo, and Gancao on the gastrointestinal motility of rats with Ganyuqizhi induced functional dyspepsia(FD). Methods: 96 Wistar rats were randomly divided into the control group,the model group, domperidone group, and three doses of Chaihushugansan groups(189、378、756 mg/kg). Chronic caudal stress and diet disorders were used to establish FD model. Gastric emptying rate, intestinal propulsive rate and gastric residential rate were observed by quantitative method. After the treatment, Ghrelin, MTL levels and VIP were measured by ELISA. The expressions of ZO-2, YAP and CGRP in rat gastric tissues were detected by Western blot. Results:Compared with the control group, the gastric emptying rate, intestinal propulsion rate, the levels of Ghrelin and MTL were significantly decreased, ZO-2 protein level was significantly down regulated, the protein levels of YAP and CGRP were significantly up regulated in the model group. Compared with the model group, the gastric emptying rate and intestinal propulsion rate were significantly increased in Chaihushugansan groups, levels of Ghrelin and MTL were increased while VIP level was decreased, ZO-2 protein level was significantly up regulated, while protein levels of YAP and CGRP were significantly down regulated.Conclusion: Chaihushugansan effective composition can effectively improve the functional dyspepsia in rats, and regulate the secretion of gastrointestinal hormones and related protein expression.
