青钱柳黄酮干预肥胖大鼠胰岛素抵抗的作用研究 被引量:11

摘要 目的:探讨青钱柳黄酮对高脂高胆固醇饮食诱导的肥胖大鼠胰岛素抵抗的影响。方法:分离并纯化青钱柳黄酮,采用LC-MS/MS方法测定指标成分含量;取肥胖敏感雄性SD大鼠给予高脂高胆固醇饲料,并随机分为青钱柳黄酮100、250、500 mg/kg组,阳性对照二甲双胍100 mg/kg组;正常对照组大鼠给予普通饲料。12周后,大鼠禁食16 h测定空腹血糖及葡萄糖耐量,称定脏器及脂肪质量;检测大鼠血清中胰岛素、ALT、AST、TC、TG水平,以及肝脏组织中MDA、SOD含量,计算胰岛素抵抗指数和血糖曲线下面积,肝、肾组织经HE染色,评价病理变化。结果:与正常组比较,模型组大鼠体重明显增加,脂肪比率增大,脏器指数、转氨酶、血脂、氧化应激等指标异常。经青钱柳黄酮干预后,剂量依赖地使肥胖模型大鼠体重明显下降,脂肪比率下降,脏器指标趋近于正常值,转氨酶、血脂、氧化应激等指标显著改善,肝脏组织脂肪空泡,炎症浸润等病理改变趋向好转。结论:青钱柳黄酮能够减轻高脂饮食诱导的肥胖大鼠体重和胰岛素抵抗,改善糖脂代谢紊乱。 Objective: To investigate the effects of Cyclocarya paliurus flavonoids(CPF) on the obesity rats, which suffer a insulin resistance induced by high-fat and high-cholesterol diet(HFD). Methods: Separation and purification of CPF were performed, LC-MS/MS method was served to determine the contents of target ingredients. Obesity-sensitive male rats were randomly divided into the model group, three doses of CPF groups, the positive control group, and rats in these groups were fed with HFD, rats in the control group were fed with ordinary feed. Fasting blood glucose and glucose tolerance were measured after fasted 16 hours at the end of the 12 weeks. Blood was taken from abdominal cavity after anesthesia, the organs and adipose tissues were immediately separated and weighed. The levels of insulin, ALT, AST, TC, TG, MDA, SOD were measured by commercial kits. Insulin resistance index and areas under the glucose curves were calculated. The pathological changes of liver and kidney tissues were evaluated by H&E staining. Results: After CPF intervention, the body weight, perirenal fat and epididymal fat pad weight of obesity rats significantly decreased, the organ index approached the normal value, the fasting blood glucose, areas under the glucose curves and insulin resistance index significantly decreased, transaminase, blood lipid, oxidative stress indexes tended to be better, pathological changes such as fat vacuoles and inflammatory infiltration in liver tissues had gradually improved. Conclusion: CPF reduce the body weight and insulin resistance of the obesity rat induced by HFD, improve the disorder condition of glucose and lipid metabolic.
