显色法检测非发酵菌对阿米卡星、哌拉西林他唑巴坦、多黏菌素B的敏感性

Susceptibility test of non-fermentative bacteria to Amikacin,Piaeracillin-tazobactam and Polymyxin B in colorimetric method

摘要 目的评估显色法检测非发酵菌对几种抗生素的药敏结果的可行性。方法采用刃天青显色法检测非发酵菌对抗菌药物阿米卡星、哌拉西林他唑巴坦、多黏菌素B的MIC值,同时选择微量肉汤法作为金标准进行评价,探讨了应用该方法的可行性。结果刃天青的适宜试验浓度为10μg/ml^20μg/ml。所测124株临床菌株,阿米卡星:EA结果为100%,CA结果为100%。哌拉西林他唑巴坦:EA结果为100%,CA结果为100%。多黏菌素B:EA结果为100%,CA结果为100%。经卡方检验,2种试验方法测该124株临床菌株MIC值差异无统计学意义(P> 0. 05)。结论刃天青显色法进行药敏试验,具有方法简便、人工目力易判读等优点,对于临床用药有一定的指导意义。 Objective To evaluate the feasibility of detecting susceptibility of non-fermentative bacteria to several antibiotics in colorimetric method. Methods Resazurin colorimetric method was used to detect minimum inhibitory concentration( MIC)value of non-fermenting bacteria to three antibiotics( Amikacin,piperacillin tazobactam,polymyxin B). At the same time,broth micro-dilution method was chosen as the gold standard to evaluate,and the feasibility of applying this method was discussed.Results The suitable concentration of resazurin was within 10 μg/ml-20 μg/ml. For 124 clinical strains,compared with the broth method,the EA of amikacin was 100% and the CA was 100%;Chi-square test showed that there was no statistical significance on the difference in MIC values of the 124 clinical strains between the two test methods. Conclusion Resazurin colorimetric method used for drug sensitivity test is simple and of easy interpretation by artificial vision. Therefore,it has certain guiding significance for clinical medication.
