摘要
目的探究结直肠癌患者血清白细胞介素37(interleukin-37,IL-37)的变化及其与肿瘤血管生成相关因子如基质金属蛋白酶-8(matrix metalloproteinase-8,MMP-8)、基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)、血管内皮生长因子(vascular endothelial growth factor,VEGF)及可溶性细胞间黏附分子(soluble intercellular adhesion molecule,sICAM-1)等的相关性。方法选取58例明确诊断的结直肠癌初诊患者,同时选取健康体检者50例作为对照组。酶联免疫吸附试验测定结直肠癌患者和对照组血清IL-37和MMP-8、MMP-9、VEGF及sICAM-1的浓度。Pearson分析IL-37与各指标间的相关性。结果结直肠癌患者血清IL-37浓度相较对照组显著下降,而MMP-8、MMP-9、VEGF及sICAM-1等均显著下调,差异均有统计学意义(P <0. 05)。IL-37、MMP-9、VEGF及sICAM-1与肿瘤大小、淋巴结转移与否及TNM分期均密切相关;MMP-8仅与淋巴结转移与否相关。此外,血清IL-37与MMP-9、VEGF及sICAM-1间呈显著负相关。结论 IL-37在结直肠癌患者血清中异常下调并与疾病严重程度相关,且与肿瘤血管生成相关因子MMP-9、VEGF及sICAM-1相关。
Objective To investigate the changes of serum IL-37 in patients with colorectal cancer( CRC) and the correlation with tumor angiogenesis-related factors such as matrix metalloproteinase-8( MMP-8),matrix metalloproteinase-9( MMP-9),vascular endothelial growth factor( VEGF) and soluble intercellular adhesion molecule( sICAM-1). Methods A total of newly diagnosed 58 CRC patients were selected. 50 healthy people were selected as control group. The levels of serum IL-37 and MMP-8,MMP-9,VEGF and sICAM-1 in CRC patients and control groups were determined by enzyme-linked immunosorbent assay. The correlations between IL-37 and each indicator were analyzed by Pearson. Results Serum IL-37 in CRC patients were significantly lower than those in control group,while MMP-8,MMP-9,VEGF and sICAM-1 were significantly down-regulated,and the differences were all statistically significant( P < 0. 05). The expressions of IL-37,MMP-9,VEGF and sICAM-1 in serum of CRC patients were correlated with tumor size,TNM stage and lymphnode metastasis( P <0. 05). With the progression of disease,their expressions were significantly decreased or increased. In addition,serum IL-37 level had a negative correlation to MMP-9,VEGF and sICAM-1. Conclusion IL-37 was down-regulated in serum of patients with CRC and was associated with disease severity and tumor angiogenesis-related factors MMP-9,VEGF and sICAM-1.
