摘要
目的:优化青刺果仁总黄酮的脱脂工艺,并对其抗氧化活性进行初步考察。方法:采用正己烷为脱脂剂,以脱油率为考察指标,用正交试验方法优化青刺果仁脱脂工艺。在最佳脱脂工艺条件下,测定脱脂后青刺果仁总黄酮含量及其抗氧化活性。结果:最佳脱脂工艺为:浸出次数5次,料液比1∶2,浸出时间100 min;在此工艺条件下,经三次试验验证,平均得油率为16.01%,脱脂后,采用柱色谱分离纯化。青刺果仁总黄酮含量为52%,对DPPH·和ABTS^+·表现出较强的清除能力,其IC50值分别为0.014 0,0.163 0 mg·ml^-1。结论:青刺果仁经最佳脱脂工艺脱脂后,总黄酮含量达50%以上,且青刺果仁总黄酮表现出很强的抗氧化能力,未来在抗氧化药物、化妆品等领域内具有一定的开发和研究价值。
Objective: To optimize the defatting method of total flavonoids from kernels of Prinsepia and investigate its antioxidant activities.Methods: With n-hexane as the defatting agent and oil yield as the index,the defatting process was optimized by orthogonal test.The content and the antioxidant activity of total flavonoids from kernels of Prinsepia were determined under the optimal conditions.Results: The optimal conditions were as follows: the leaching times was five,the solid-liquid ratio was 1∶2,and the leaching time was 100 min.The average oil yield was 16.01% under the optimal conditions.The yield of flavonoids was 52% after the purification,meanwhile,the flavonoids from kernels of Prinsepia showed strong DPPH· and ABTS^+· radical scavenging activities with IC50 value of 0.014 0 mg·ml^-1 and 0.163 0 mg·ml^-1,respectively.Conclusion: The content of flavonoids is over 50% after being purified under the optimal conditions,and the flavonoids show strong DPPH· and ABTS+· radical scavenging activities.Therefore,the flavonoids from kernels of Prinsepia have superior research value in antioxidant drugs,cosmetics and other fields.
