摘要
目的:体外实验研究TGF-β1诱导人膀胱癌细胞T24发生上皮-间质转化(EMT)的作用及其机制。方法:以人膀胱癌细胞T24作为研究对象,使用不同浓度TGF-β1干预24 h,光镜下观察T24细胞形态学变化;分别采用Real time-PCR(RT-PCR)和Western blot法检测细胞中上皮表型蛋白E-cadherin和间质表型蛋白Vimentin mRNA和总蛋白的表达。Transwell小室实验检测细胞的侵袭与迁移潜能。结果:TGF-β1可激活T24细胞的EMT程序,并上调Vimentin mRNA和蛋白的表达水平和下调E-cadherin mRNA和蛋白的表达水平。此外,TGF-β1可促进T24细胞体外侵袭潜能,而TGF-β1拮抗剂LY2109761可阻断这一过程。结论:EMT过程的激活参与了TGF-β1介导的人膀胱癌T24细胞的侵袭。
Objective:To investigate the role of TGF-β1 on epithelial mesenchymal transition(EMT) and invasion ability in human bladder cancer cell line T24.Methods:Cultured T24 cells were pre-treated with different concentrations of TGF-β1 for 24 h.The morphological changes were observed under phase-contrast microscopy.The mRNA and protein expression levels of EMT relative marker E-cadherin and Vimentin were detected by RT-PCR and Western blot.The effect of TGF-β1 on migration and invasion abilities of T24 cell line was detected with transwell migration and invasion assay.Results:Our present results demonstrated that TGF-β1 could induce T24 morphological change from epithelial to mesenchymal.The mRNA and protein level of epithelial marker E-cadherin was downregulated by TGF-β1,whereas the mRNA and protein expression level of mesenchymal maker protein Vimentin was upregulated.Furthermore,TGF-β1 significantly enhances the migration and invasion ability of T24 cells,however TGF-β1 inhibitor LY2109761 could effectively reverse this process.Conclusion:TGF-β1 enhances T24 cells migration and invasion by inducing epithelial mesenchymal transition.
