1株产菊粉酶菌株的筛选及发酵条件优化

摘要 研究以透明圈法从菊芋根际土壤中筛选产菊粉酶菌株,通过初筛和复筛,筛选出1株产菊粉酶活力较高的菌株Z-4,经菌落和显微镜观察,初步鉴定为霉菌。通过正交试验优化菌株Z-4的发酵条件,分析不同因素对该菌株产菊粉酶活力的影响,结果表明,菌株Z-4产菊粉酶的最优条件为:以菊粉为唯一碳源,最佳氮源为酵母浸粉、添加量9g/L,培养温度32℃,初始pH6.5,培养时间7d,此时菌株Z-4实际产酶活力达到15.62U/mL。 This study screened the inulinase-producing strain from the rhizosphere soil of Jerusalem artichoke with transparent circle method.A strain Z-4 with high enzyme activity was screened by primary and secondary screening by.A strain Z-4 initially identified as mold after colony morphology and microscopic observation.Analysis the effects of different single factors on the inulinase activity.The optimization of fermentation conditions for Z-4 strain by orthogonal experimental,The results show that the optimal condition was: inulin as the sole carbon source,The best nitrogen source was yeast extract、added amount 9g/L,culture temperature 32℃,initial pH6.5,culture time 6d,under this condition,the enzyme activity of Z-4 strain reached 15.62U/mL.
