摘要
目的研究平喘Ⅰ号调节基质细胞衍生因子-1(SDF-1)及其受体CXCR4在支气管哮喘小鼠肺内表达变化及影响。方法 72只雌性BALB/C小鼠按数字随机分为正常对照组、模型对照组、布地奈德组、平喘Ⅰ号低剂量组、中剂量组、高剂量组,共6组,每组12只。通过卵清蛋白(OVA)致敏和激发复制支气管小鼠哮喘模型,然后分别予生理盐水、布地奈德药物及中药平喘Ⅰ号低、中、高剂量干预。取小鼠肺组织,通过病理切片观察,图像分析软件测定支气管壁厚度(Wat)和平滑肌厚度(Wam),采用实时定量PCR测定小鼠肺组织中SDF-1 mRNA及CXCR4 m RNA的含量,通过免疫印迹法检测肺组织中SDF-1及CXCR4的蛋白表达水平,采用线性相关分析技术测量了SDF-1与CXCR4的相关性。结果 SDF-1及CXCR4的表达水平在正常对照组中较低,在模型对照组中表达水平明显增高(P均<0.01)。与模型对照组比较,平喘Ⅰ号各剂量组和布地奈德组中SDF-1及CXCR4的表达水平均明显降低(P<0.01或P<0.05);其中布地奈德组表达水平低于平喘Ⅰ号低剂量和中剂量组(P<0.05或P<0.01),而中药高剂量组的表达水平低于布地奈德组(P<0.05)。结论在支气管哮喘气道炎症及其气道重塑过程中,SDF-1及其受体CXCR4两者相互作用,干预其重塑过程。平喘Ⅰ号通过影响SDF-1及CXCR4在肺内的表达而干预哮喘气道重塑,达到缓解哮喘症状的作用。
Objective To study the influence of Pingchuan Ⅰ in regulating stromal cell-derived factor-1 (SDF-1) expression and its receptor CXCR4 in the lung of bronchial asthma mice.Methods 72 female BALB/C mice were randomly divided into normal control group,model control group,budesonide group,Pingchuan Ⅰ low-dose group,middle-dose group and high-dose group.There were 12 mice in each group.The bronchial mouse asthma model was copied by ovalbumin (OVA) sensitization and stimulation,and then administered with normal saline,budesonide and low,medium and high doses of Chinese medicine Pingchuan Ⅰ.Mouse lung tissues were taken and observed by pathological sections.Image analysis software was used to measure bronchial wall thickness (Wat) and smooth muscle thickness (Wam).Real-time quantitative PCR was used to determine SDF-1 mRNA and CXCR4 mRNA in mouse lung tissues.The expression levels of SDF-1 and CXCR4 in lung tissue were detected by Western blotting.The correlation between SDF-1 and CXCR4 was measured by linear correlation analysis.Results The expression levels of SDF-1 and CXCR4 were low in the normal control group,and the expression levels in the model control group were significantly higher (P<0.01).Compared with those of the model control group,the expression levels of SDF-1 and CXCR4 in the Pingchuan I each dose group and the budesonide group were significantly lower(P<0.01 or P<0.05).The expression level in budesonide group was lower than that in the low-dose and medium-dose Pingchuan Ⅰ group (P<0.05 or P<0.01).While the expression level of the high dose Chinese medicine group was lower than that of the budesonide group(P<0.05).Conclusion In the process of airway inflammation and airway remodeling in bronchial asthma,SDF-1 and its receptor CXCR4 are involved in the remodeling process.Pingchuan Ⅰ interferes with airway remodeling in asthma by affecting the expression of SDF-1 and CXCR4 in the lungs,to relieve the symptoms of asthma.
