摘要
目的 检测细胞周期与细胞凋亡调控蛋白(CCAR)1mRNA、蛋白在胃腺癌组织中表达及其临床意义。方法 通过免疫组化链霉菌抗生物素蛋白-过氧化物酶连结法(SP)法、Western印迹法和实时荧光定量-聚合酶链反应(qRT-PCR)技术检测60例胃腺癌组织及40例癌旁正常胃黏膜组织中CCAR1mRNA、蛋白表达。结果 胃腺癌组织中CCAR1mRNA、蛋白的表达量较癌旁正常胃黏膜组织明显升高(P<0.05),表达水平随肿瘤分化程度的降低而显著增加(P<0.05)。结论 随分化程度的降低CCAR1mRNA、蛋白的表达量增加,其表达的异常升高与胃腺癌的发生密切相关。
Objective To observe the expression of cell cycle and apoptotic regulatory protein (CCAR)1 in gastric adenocarcinoma tissues and explore its clinical significance. Methods Immunohistochemical SP method, Western blot and qRT-PCR were used to detect the expression of CCAR1 protein and mRNA in 60 cases of gastric adenocarcinoma and 40 cases of normal gastric mucosa adjacent to cancer. The role of CCAR1 in the occurrence and development of gastric adenocarcinoma and its relationship with clinicopathological characteristics were analyzed. Results The expressions of CCAR1 mRNA and protein in gastric adenocarcinoma were significantly higher than those in normal gastric mucosa adjacent to cancer ( P <0.05). The expressions of CCAR1 mRNA and protein in gastric adenocarcinoma tissues were related to the degree of differentiation ( P <0.05). Conclusions The abnormal high expression of CCAR1 is closely related to the occurrence of gastric adenocarcinoma.
