类固醇激素受体起始密码子区突变位点报告基因系统的构建 被引量:1

摘要 目的 构建含有类固醇激素受体(VDR)起始密码子区突变序列的双荧光素酶报告基因系统。方法 以人肝组织总RNA为模板应用RT-PCR方法扩增出VDRcDNA基因序列全长并克隆于T载体后酶切再与空载体pcDNA3.1(-)B-myc/his连接形成重组载体;测序验证后与荧光素酶报告基因连接构建起始密码子区T/C突变型人VDR,以phRL-SV40质粒作为内对照共转染COS-7细胞,设1,25(OH)2VD3不同浓度点进行干预后,化学发光法检测荧光强度。结果 人VDR基因起始密码子区突变重组质粒测序验证与Genbank完全相同。通过双荧光素酶报告基因分析系统检测,1,25(OH)2VD3干预组荧光素酶活性数值显著升高,差异有统计学意义(P<0.05),表明人VDR起始密码子区突变重组体在COS-7细胞中存在转录激活能力。结论 成功构建了含有起始密码子区突变的报告基因载体,为VDR有关的药物代谢异常、肿瘤易感性提供机制性指导。
