超高效液相色谱-串联质谱法同时测定生物体液中的酪氨酸及其代谢产物 被引量:2

Simultaneous Determination of Tyrosine and Its Metabolites in Biological Fluids by Ultra-high Performance Liquid Chromatography-Tandem Mass Spectrometry

摘要 本文采用Shim-pack GIST C18色谱柱,以20 mmol/L的HAc-NH 4Ac缓冲溶液(pH=5.7)∶乙腈=95∶5(V/V)为流动相,建立了超高效液相色谱-串联质谱法(UPLC-MS/MS)同时测定大鼠尿液中酪氨酸及其代谢产物的分析方法。在优化的实验条件下,分析物线性关系良好(R^2>0.9991),检出限在0.0003~0.0041 mg/L之间,3个浓度水平的加标回收率范围为82.0%~113.0%,相对标准偏差(RSD)小于4.4%。该方法简单、快速、灵敏度高,可用于尿液中酪氨酸及其代谢产物的分析测定。 A new analytical method was developed for the simultaneous determination of tyrosine and its metabolites by ultra-high performance liquid chromatography-tandem mass spectrometry.With isocratic elution using acetic acid-ammonium acetate(20 mmol/L,pH=5.7)and acetonitrile(95∶5,V/V),tyrosine and its metabolites were well separated on Shim-pack GIST C18 column.Under the optimized conditions,the linear relationships for tyrosine and its metabolites were good(R^2>0.9991),and the limits of detection were between 0.0003 and 0.0041 mg/L.Recoveries of tyrosine and its metabolites in rat urine were in the range of 82.0%-113.0%,with relative standerd deviations less than 4.4%.The present method was simple,rapid and sensitive,which was successfully used for the simultaneous determination of tyrosine and its metabolites in urine.
