星点设计-效应面法优化叶黄素脂质体的处方及制备工艺 被引量:6

摘要 目的:采用星点设计-效应面法优化叶黄素脂质体的处方和制备工艺。方法:在单因素试验基础上,以包封率、粒径和渗漏率为自变量,叶黄素用量、大豆卵磷脂与胆固醇质量比、旋蒸温度为因变量,利用星点设计模型进行二项式拟合,通过三维效应面图和等高线图选择最佳处方,并对预测结果进行验证。结果:叶黄素脂质体的最佳处方和制备工艺为叶黄素用量5.34mg,大豆卵磷脂与胆固醇质量比为14.35∶1,旋蒸温度为32.9℃。在该条件下,3批叶黄素脂质体样品的平均包封率为(87.49±0.33)%,粒径为(68.52±0.21)nm,渗漏率为(14.54±0.50)%。结论:用星点设计-效应面法优化的叶黄素脂质体的制备工艺简便、稳定、可行,适用于叶黄素脂质体的制备。 Objective: To optimize the formulation and preparation technology of lutein liposomes by central composite design-response surface method.Methods:On the basis of single factor test,entrapment efficiency,particle size and leakage rate were selected as independent variables,lutein dosage,mass ratio of soybean lecithin to cholesterol,rotary steaming temperature were selected as dependent variables.The results were fitted by design-response surface methodology.The formulation was optimized by three-dimensional response surface and contour lines graphs.The predictive data were validated.Results:The optimal formulation and preparation technology were as follows:the lutein dosage was 5.34 mg,mass ratio of soybean lecithin to cholesterol was 14.35∶1,rotary steaming temperature was 32.9 ℃.Under the optimal conditions,the average entrapment efficiency of three batches of lutein liposomes was (87.49±0.33)%,the particle size was (68.52±0.21) nm and the leakage rate was (14.54±0.50)%.Conclusion:The preparation technology of lutein liposomes by central composite design-response surface method is simple,stable and feasible,and is suitable for the preparation of lutein liposomes.
