p53及p16在皮肤鳞状细胞癌和鲍温病中的表达及意义 被引量:4

Expression and Significance of p53 and p16 Protein in Cutaneous Squamous Cell Carcinoma and Bowen’s Disease

摘要 目的通过检测皮肤鳞状细胞癌(CSCC)和鲍温病(BD)组织中p53和p16蛋白的表达情况,分析p53和p16蛋白表达之间的相互关系,探讨二者在CSCC和BD发生、发展中所起的作用。方法应用免疫组织化学SP法检测50例皮肤鳞癌组织(其中高分化鳞癌30例,中低分化鳞癌20例)、30例BD组织以及20例正常皮肤组织标本中p53和p16蛋白的表达及分布。结果p53蛋白在正常皮肤组织、BD及CSCC中表达的阳性率分别10.00%、36.67%及80.00%,p53蛋白在高分化鳞癌和中低分化鳞癌中表达的阳性率分别为76.67%和85.00%,p53蛋白在BD和CSCC中表达阳性率均高于正常皮肤组织(P<0.05)。p53蛋白在BD中表达阳性率低于CSCC,二者差异有显著统计学意义(P<0.001)。p53蛋白在BD、高分化鳞癌及中低分化鳞癌中的表达率呈上升趋势,差异有显著统计学意义(P<0.001)。p16蛋白在正常皮肤组织、BD及皮肤CSCC中表达的阳性率分别为0.00%、53.33%及22.00%,在高分化鳞癌和中低分化鳞癌中表达的阳性率分别为30.00%和10.00%。p16蛋白在BD和CSCC中表达阳性率均高于正常皮肤组织(P<0.05)。p16蛋白在BD中表达阳性率高于肾CSCC,二者间差异有统计学意义(P<0.05)。p16蛋白在BD、高分化鳞癌、中低分化鳞癌中的表达阳性率呈下降趋势,差异有统计学意义(P<0.05)。BD中p53和p16蛋白的表达呈正相关性(rs=0.402,P<0.05)。CSCC中p53和p16蛋白的表达呈负相关性(rs=-0.579,P<0.001)。结论p53及p16的高表达可能在BD的发生中起重要作用,且p53的高表达可能在CSCC的发生、发展及浸润中同样发挥着重要作用,p16的低表达可能对CSCC的进展有重要意义。 Objective In order to discuss the role of p53 and p16 protein in the occurrence and development of cutaneous squamous cell carcinoma (CSCC) and Bowen's disease (BD). The expression of p53 and p16 protein is detected and the relationship between them is analyzed. Methods Using immunohistochemical SP method to detect the expression and distribution of p53 and p16 protein in 50 cases of CSCC (high differentiated squamous carcinoma 30 cases,middle and low differentiated squamous carcinoma 20 cases),30 cases of BD and 20 cases of normal skin tissue. Results The positive rates of p53 protein in normal skin tissue,BD and CSCC were 10.00%,36.67% and 80.00%,respectively. The positive rate of p53 protein in high differentiated squamous carcinoma,middle and low differentiated squamous carcinoma were 76.67% and 85%. The expression positive rates of p53 protein in BD and CSCC group were significantly higher than that in normal skin tissues (P<0.05). The expression positive rate of p53 protein in BD was lower than that of CSCC,and the difference was statistically significant (P <0.001). The expression positive rate of p53 protein in BD,high differentiated CSCC,middle and low differentiated CSCC showed an upward trend,and the difference was statistically significant (P<0.001). The positive rate of p16 protein in normal skin tissue,BD and CSCC were 0.00%,53.33% and 22.00%,respectively. The positive rate of p16 protein in the high differentiated squamous carcinoma,middle and low differentiated squamous carcinoma were 30.00% and 10.00%. The expression positive rates of p16 protein in BD and CSCC group were significantly higher than normal skin tissues (P <0.05). The expression positive rate of p16 protein in BD was higher than that of CSCC,and the differences were statistically significant (P <0.05). The expression positive rate of p16 protein in BD,high differentiated CSCC,middle and low differentiated CSCC was on the decline,and the difference was statistically significant (P<0.05). The expression of p53 and p16 protein in BD showed a positive correlation (rs=0.402,P<0.05). The expression of p53 and p16 protein in CSCC showed a negative correlation (rs =-0.579,P <0.001). Conclusion High expression of p53 and p16 may play an important role in the occurrence of BD. High expression of p53 also may play an important role in the occurrence,development and infiltration of CSCC. Low expression of p16 may be important for the development of CSCC.
