摘要
目的制备新型纳米给药系统槲皮素@介孔羟基磷灰石(QUE@MHAs),对其进行表征,并研究其体外释放度。方法采用透射电镜,动态光散射仪和Zeta电位分析仪对纳米材料进行表征,通过高效液相色谱法在体外考察纳米粒的释药行为。结果QUE@MHAs的TEM平均粒径为(173.8±5.1)nm,Zeta电位为(-17.1±3.3)mV,负载量为28.3%。稳定性实验表明纳米粒的粒径基本没有变化。通过HPLC法测定QUE浓度,色谱峰的峰形较好,线性关系和精密度良好,符合要求。2h内,QUE@MHAs中的QUE的累积释放量为59.7%。结论本文制备的QUE@MHAs粒径均一,具有较好的稳定性,并能减缓QUE的释放。
Objective To prepare and characterize a new nano-drug delivery system (quercetin@mesoporous hydroxyapatite,QUE@MHAs) and investigate its release in vitro. Methods Transmission electron microscopy,dynamic light scatterometer and Zeta potential analyzer characterized QUE@MHAs.The drug release behavior of nanoparticles was investigated by HPLC. Results The average TEM particle size of QUE@MHAs was (173.8 ± 5.1)nm,the Zeta potential was (-17.1 ± 3.3)mV,and the drug load was 28.3%.The stability experiments showed that the size of nanoparticles did not change.QUE was determined by HPLC,the peak shape of chromatographic peak was good,and the linear relationship and precision were good.Within 2 hours,the cumulative release amount of QUE in QUE@MHAs is 59.7%. Conclusion The nanoparticles (QUE@MHAs) prepared in this paper have uniform particle size,good stability,and slow release effect.
