摘要
目的研究环状RNA(circRNA)hsa_circ_0010985在胃癌(GC)细胞和组织中的表达,并探讨hsa_circ_0010985对GC细胞生物学功能的影响及其临床意义。方法实时荧光定量PCR(qRT-PCR)检测hsa_circ_0010985在GC细胞和组织中的表达情况,并分析hsa_circ_0010985在GC中的表达及其与临床病理特征的关系,通过受试者工作曲线(ROC曲线)探讨hsa_circ_0010985在GC中的诊断价值;利用小干扰RNA (siRNA)转染GC细胞BGC823,以敲低hsa_circ_0010985的表达,通过细胞计数方法(CCK-8)检测hsa_circ_0010985对GC细胞增殖的影响;流式细胞仪检测hsa_circ_0010985对GC细胞凋亡的影响。结果 hsa_circ_0010985在GC细胞和组织中高表达,hsa_circ_0010985在GC组织中的表达水平显著高于癌旁正常组织,差异有统计学意义( P <0.05),且与淋巴结转移之间差异有统计学意义( P <0.05),而与患者性别、年龄、肿瘤直径、TNM分期、癌胚抗原(CEA)、糖类抗原CA199(CA199)、肿瘤位置之间差异无统计学意义( P >0.05);通过ROC曲线分析,ROC曲线下面积(AUC)为0.631( P <0.05),特异度和灵敏度分别为0.814和0.419,提示其可能对GC的诊断具有一定的参考价值;敲低hsa_circ_0010985促使GC细胞增殖能力减弱,凋亡率增加。结论 hsa_circ_0010985可能促进GC细胞发生发展,并有望成为GC诊断的生物标志物。
Objective To study the expression of hsacirc0010985 in gastric cancer(GC) cells and gastric tissues, and explore the effects of hsacirc0010985 expression on the proliferation and apoptosis of GC cells and its clinical significance. Methods Quantitative polymerase chain reaction assay(qRT-PCR) was performed to detect the expression level of hsacirc0010985 in GC cells and tissues. And the relationship between the expression level of hsacirc0010985 and the clinicopathological parameters of GC patients was analyzed. A receiver operating characteristic(ROC) curve was used to evaluate the diagnostic value of hsacirc0010985. BGC-823 cells were transfected with siRNA to knockdown hsacirc0010985 expression. And then, cell proliferation and apoptosis were detected by CCK8 and AV/PI assays. Results Hsacirc0010985 was highly expressed in gastric cancer cells and tissues(P<0.05). Its expression levels were significantly correlated with lymphatic metastasis(P<0.05), but there were no significant differences between gender, age, tumor size, TNM stage, CEA, CA199 and tumor location(P<0.05). The areas under the ROC curve(AUC) of hsacirc0010985 in tissues were up to 0.631. The specificity and sensitivity were 0.814 and 0.419;Decreased hsacirc0010985 expression suppressed the proliferation and promoted apoptosis of GC cells. Conclusion Hsacirc0010985 may serve as a novel tumor promoter and a potential biomarker in GC.
