摘要
本文讨论了一种可应用于大部分液相色谱系统延迟体积的测算方法。本次测算的对象是北京卫星制造厂在科技部重大专项《高精度超高压液相泵的开发与应用》(项目编号:2016YFF0101000)项目中研制的高精度超高压液相泵(UHPLC-BSC-02型),以及SCG蛋白纯化系统中的色谱泵。该方法配合赛谱仪器UV检测器使用,是一种方便可靠的测算方法,测算对象灵活,应用范围广且具有良好的重复性。根据实际情况,可以用于大多数液相系统的延迟体积、泵死体积、样品泵加载体积、柱外死体积等测量。
In this paper,a method for measuring the delay volume of most liquid chromatography systems is discussed.The object of this measurement is the high precision ultra-high pressure chromatography pump (UHPLC-BSC-02)developed by Beijing satellite manufacturing co.LTD,and a chromatography pump in SCG system.This method is a convenient and reliable measurement method,which need to be used in combination with UV detector of Sepure Instrument co.LTD.It has flexible measurement objects,wide application range and good repeatability.According to the actual situation,it can be used to measure gradient delay volume,pump dead volume,sample pump loading volume,and dead volume outside column in most liquid chromatography systems.
