温畅祛痛汤对寒凝痛经模型大鼠的影响 被引量:1

摘要 目的 观察温畅祛痛汤对寒凝痛经模型大鼠的影响。方法 将60只SD大鼠随机分为空白对照组10只及造模组50只。造模组采用皮下注射苯甲酸雌二醇加寒冷刺激制备大鼠痛经模型,并按随机数字表法分为模型组、阳性药组及温畅祛痛汤高、中、低剂量组,每组10只。阳性药组予痛经宝颗粒混悬液20 g/kg灌胃,温畅祛痛汤高、中、低剂量组分别予温畅祛痛汤24、12、6 g/kg灌胃,空白对照组、模型组予等容积蒸馏水灌胃,每日1次,每次10 mL/kg,共10 d。观察各组大鼠扭体反应,测量大鼠子宫收缩情况,检测子宫组织中6-酮-前列腺素F 1α(6-keto-PGF 1α)、血栓烷B 2(TXB 2)、一氧化氮(NO)及内皮素(ET)含量,采用实时荧光定量聚合酶链式反应(Real-time RT-PCR)法检测子宫组织中内皮型一氧化氮合酶(eNOS)、诱导型一氧化氮合酶(iNOS)mRNA表达。结果 温畅祛痛汤能显著延长寒凝痛经模型大鼠的扭体潜伏期( P < 0.01 ),减少扭体次数( P <0.05),减少子宫收缩波数量( P <0.01, P <0.05)和收缩峰峰值( P <0.05),抑制子宫活动度( P <0.01),升高子宫组织中6-keto-PGF 1α和NO含量( P <0.01, P <0.05),降低TXB 2和ET含量( P <0.01, P <0.05),上调子宫组织中eNOS mRNA表达( P <0.01, P <0.05),下调iNOS mRNA表达( P < 0.01 )。以温畅祛痛汤高剂量组作用最为明显。结论 温畅祛痛汤有良好的抗痛经作用,且呈剂量依赖性,以温畅祛痛汤高剂量组效果最佳,其作用机制可能与抑制子宫收缩,扩张血管,缓解子宫缺血乏氧状态有关。 Objective To observe the effect of Wenchang Qutong decoction on cold coagulation dysmenorrhea model rats. Methods 60 SD rats were randomly divided into blank control group (10 rats) and model group (50 rats). Rat dysmenorrhea model was established by subcutaneous injection of estradiol benzoate and cold stimulation in the model group. The rats were divided into model group, positive drug group and Wenchang Qutong decoction high, medium and low dose groups according to random number table method, with 10 rats in each group. The positive drug group was given Tongjingbao granule suspension 20 g/kg by gavage, the high, medium and low dose groups were given Wenchang Qutong decoction 24, 12, 6 g/kg respectively by gavage, while the blank control group and model group were given isovolumetric distilled water by gavage once a day, 10 mL/kg each time for 10 days. The writhing reaction and uterine contraction of rats in each group were observed. The contents of 6-keto-prostaglandin F 1α(6-keto-PGF 1α), thromboxane B 2 (TXB 2), nitric oxide (NO) and endothelin (ET) in uterine tissues were measured. Real-time quantitative polymerase chain reaction (Real-time RT-PCR) was used to detect the mRNA expression of endothelial nitric oxide synthase (eNOS) and inducible nitric oxide synthase (iNOS) in uterine tissues. Results Wenchang Qutong decoction can significantly prolong the writhing latency of rats with cold coagulation dysmenorrhea model ( P <0.01), reduce writhing times ( P <0.05), reduce the number of uterine contraction wave ( P <0.01 , P <0.05) and peak value of uterine contraction wave ( P <0.05), inhibit uterine activity ( P <0.01), increased 6-keto-PGF 1α and NO content in uterine tissue ( P <0.01, P <0.05), and decreased TXB 2 and ET content ( P <0.01, P <0.05), up-regulated the expression of eNOS mRNA in uterine tissue ( P <0.01, P <0.05), and down-regulated the expression of iNOS mRNA ( P <0.01). The effect of Wenchang Qutong decoction high dose group was the most obvious. Conclusion Wenchang Qutong decoction has a good anti-dysmenorrhea effect in a dose-dependent manner.The high dose group of Wenchang Qutong decoction has the best effect. Its mechanism may be related to inhibiting uterine contraction, dilating blood vessels and alleviating hypoxic and ischemic state of uterus.
