摘要
磷脂酶A2 (phospholipaseA2, PLA2)是一种能催化磷脂甘油分子上二位酰基的水解酶。蓖麻PLA2α是参与酰基编辑处理过程中的一个关键酶。根据NCBI上已知蓖麻PLA2α基因(LOC8289506)及其基因组(NW-002994322)序列设计引物,PCR扩增得到PLA2α基因启动子片段。生物信息学分析PLA2α基因及启动子序列。结果表明克隆到PLA2α基因启动子序列2 412 bp,分析预测显示有多个转录起始位点及大量的顺式作用元件,PLA2α基因共编码160个氨基酸,存在跨膜结构,并用MEGA 7.0构建了进化树,发现蓖麻和麻疯树,巴西橡胶树,木署聚类到一起,这一分类与生物学分类一致,均为大戟科植物。对蓖麻PLA2α基因及其启动子的研究,为蓖麻胚乳中脂肪酸的合成和TAG积累的研究提供理论和实践基础。
Phospholipase A2 (PL2) is a hydrolase that catalyzes the diacyl group on the phospholipid glycerol molecule. Castor PLA2α is a key enzyme involved in the acyl editing process. Primers were designed based on the known castor PLA2α gene (LOC8289506) and its genome (NW-002994322) sequences on NCBI, and the PLA2α gene promoter fragment was obtained by PCR amplification. PLA2α gene and promoter sequences were analized by bioinformatics. The results showed that the promoter sequence of PLA2α gene was 2 412 bp, and it was predicted to have multiple transcription initiation sites and a large number of cis-acting elements. The PLA2α gene encoded 160 amino acids with transmembrane structure, it was secretory protein, and constructed a phylogenetic tree by MEGA 7.0, to found that the castor and jatropha, the Brazilian rubber tree, and the grove were clustered together. This classification was consistent with the biological classification, and all of them were Euphorbiaceae plants. This experiment mainly studied the castor PLA2α gene and its promoter, and laid theoretical and practical basi for the study of fatty acid synthesis and TAG accumulation in castor endosperm.
