怀玉山高山马铃薯不同脱毒方法效果比较 被引量:1

Comparison of the Effects of Different Virus-Free Methods on Huaiyushan Alpine Potato

摘要 为了找出怀玉山高山马铃薯一种较好的脱毒方法,本研究对怀玉山高山马铃薯茎尖再生苗(常规茎尖培养,热处理+茎尖培养,茎尖玻璃化法超低温疗法,茎尖包埋玻璃化法超低温疗法以及茎尖小滴玻璃化法超低温疗法)的脱毒效果进行RT-PCR检测和比较。结果表明,对于PVA来说,常规茎尖脱毒效果较差,而包埋玻璃化法超低温疗法茎尖脱毒效果最好;对于PVM来说,热处理+茎尖脱毒、玻璃化法超低温疗法茎尖脱毒、包埋玻璃化法超低温疗法茎尖脱毒以及小滴玻璃化法超低温疗法茎尖脱毒均可达到较好的效果;对于PVS来说,常规茎尖脱毒、热处理+茎尖脱毒和包埋玻璃化法超低温疗法茎尖脱毒的效果较好;对于PLRV来说,常规茎尖脱毒和包埋玻璃化法超低温疗法茎尖脱毒的效果较好;对于PVX来说,热处理+茎尖脱毒、玻璃化法超低温疗法茎尖脱毒、包埋玻璃化法超低温疗法茎尖脱毒和小滴玻璃化法超低温疗法茎尖脱毒均可获得较好的脱毒效果;对于PVY来说,包埋玻璃化法超低温疗法茎尖脱毒的效果最佳。包埋玻璃化法超低温疗法茎尖脱毒是怀玉山高山马铃薯脱毒较为适宜的脱毒方法。本实验结果可为怀玉山高山马铃薯脱毒苗的培育提供一些技术参考。 In order to find a better virus-free method for alpine potato in Huaiushan, the virus-free effects of plantlets regenerated from shoot-tips by 5 kinds of virus-free methods (conventional shoot tip virus-free, heat treatment and shoot tip virus-free, virus-free by vitrification cryotherapy, virus-free by encapsulation vitrification cryotherapy and virus-free by droplet vitrification cryotherapy) were detected and compared by RT-PCR. The results showed that, for PVA, the effect of conventional shoot tips virus-free was poor, and the effect of shoot tips virus-free by encapsulation vitrification cryotherapy was the best. For PVM, the heat treatment+shoot tip virus-free, virus-free by vitrification cryotherapy, virus-free by encapsulation vitrification cryotherapy and virus-free by droplet vitrification cryotherapy could all achieve better effects. For PVS, the conventional shoot tip virus-free, heat treatment +shoot tip virus-free and virus-free by encapsulation vitrification cryotherapy had better effects. For PLRV, the effects of the conventional shoot tip virus-free and virus-free by encapsulation vitrification cryotherapy were better. For PVX, the heat treatment+shoot tip virus-free, virus-free by vitrification cryotherapy, virus-free by encapsulation vitrification cryotherapy and virus-free by droplet vitrification cryotherapy could all achieve a better result. For PVY, the effect of shoot tips virus-free by encapsulation vitrification cryotherapy was the best. Virus-free of shoot tips by encapsulation vitrification cryotherapy was a more suitable virus-free method for the virus-free of Huaiyushan alpine potato. The results of the experiment might provide some technical references for the cultivation of virus-free plantlets of Huaiyushan alpine potato.
