摘要
目的建立一种同时检测人常染色体插入缺失多态性(Deletion/Insertion Polymorphisms,DIP)位点和短串联重复序列多态性(Short Tandem Repeat,STR)基因座的多重荧光标记复合扩增体系,为不均衡混合样本DNA检测提供有效的辅助方法。方法在多色荧光标记复合扩增和毛细管电泳技术的基础上,利用DIP和STR两种遗传标记串联组合成新型遗传标记DIP-STR,构建荧光标记复合扩增体系同步检测分析样本的8个DIP-STR基因座。结果建立了一种同时检测8个DIP-STR基因座分型的检测方法。结论本研究通过设计针对DIP位点插入或缺失的特异性引物,在现有法医物证常规检验设备和方法的基础上,可对现场物证中不均衡混合样本进行分型,为现场案件混合DNA检测提供有效辅助方法。
Objective To establish a fluorescence labeling multiplex amplification system for simultaneous detection of human autosomal Deletion/Insertion Polymorphisms (DIP) and Short Tandem Repeat (STR) loci,which provides an effective auxiliary method for forensic mixed sample detection.Methods On the basis of multi-fluorescent marker amplification and capillary electrophoresis (CE),DIP loci and STR loci were combined as DIP-STR and eight DIP-STR locus were detected and analyzed.Results A method for simultaneous detection of 8 DIP-STR locus were established.Conclusion In this study,we designed specific primers for insertion or deletion of DIP loci.On the basis of the existing forensic evidence routine testing equipment and methods,we can classify the mixed imbalanced samples in the field of physical evidence,which becomes an effective auxiliary method for case DNA detection.
