摘要 对男性、女性的外周血淋巴细胞进行染色体核型分析发现,在"男1"、"女2"视野中均可找到分散较好、形态较清晰且数量为46条的淋巴母细胞染色体。介绍了不同秋水仙素终浓度(0.3μg/ml、0.6μg/ml)对染色体中期分裂相的影响,认为这两种终浓度的秋水仙素均适合应用到人外周血淋巴细胞染色体的制备过程当中。找出了分裂相数量较少的原因,如PHA浓度、秋水仙素的细胞毒性、滴片的高度或力度不合适。要多考虑秋水仙素浓度、秋水仙素处理时间这两个指标,进而找出染色体制备的最优因素水平组合。 Karyotype analysis of peripheral blood lymphocyte of male and female showed that 46 lymphoblastic chromosomes with good dispersion,clear morphology and quantity could be found in the field of "male 1" and "female 2".The effects of different final concentrations of colchicine (0.3 ug/ml,0.6 ug/ml) on metaphase division of chromosomes were introduced.It was considered that both final concentrations of colchicine were suitable for chromosome preparation of human peripheral blood lymphocyte.The reasons for the small number of cleavage phases,such as the concentration of PHA,cytotoxicity of colchicine,the inappropriate height or strength of the droplets,should be considered more colchicine concentration and colchicine treatment time,so as to find the optimal combination of factors for chromosome preparation.
