桦褐孔菌质量标准的研究

Study on Quality Standard of Inonotus Obliquus(Fr.) Pilat

目的:建立桦褐孔菌的质量标准,为其质量控制提供参考。方法采用显微鉴别及理化鉴别对从不同产地收集的12批桦褐孔菌的进行定性鉴别;按2015年版《中国药典》(四部)通则方法测定桦褐孔菌粉样品中水分、总灰分、酸不溶性灰分含量和水溶性浸出物含量;采用分光光度法法对桦褐孔菌中的多糖含量进行测定。结果从不同产地收集的12批桦褐孔菌的鉴别结果与《山东省中药材标准》中的桦褐孔菌一致;无水葡萄糖线性范围为24~144μg,r=0.997,平均回收率为92.7%,RSD=0.7%(n=6);12批样品中水分、总灰分、酸不溶性灰分、水溶性浸出物的平均值分别为10.5%、13.8%、0.65%、30.2%;多糖平均含量为4.3%。结论:该试验所建标准可用于桦褐孔菌的质量控制。

OBJECTIVE: To establish the quality standard of Inonotus obliquus(Fr.) Pilat, and to provide reference for its quality control. METHODS: The microscopical and physicochemical identification were used to identify 12 batches of Inonotus obliquus(Fr.) Pilat from different areas. Determination of water content, total ash content, acid-insoluble ash content and water-soluble Leach content in the samples of Inonotus obliquus(Fr.) Pilat according to the method of the fourth part of the 2015 edition of the Chinese Pharmacopoeia. Spectrophotometric method was used to determine the content of polysaccharide in Inonotus obliquus(Fr.) Pilat .RESULTS:The identification results of 12 batches of Inonotus obliquus(Fr.) Pilat collected from different origins are consistent with those of birch Inonotus obliquus(Fr.) Pilat in the Shandong Provincial Chinese Herbal Medicine Standard. Anhydrous glucose linear range of 24~144 μg, R = 0.997, average recovery rate of 92.7 %, RSD = 0.7 %(N = 6);The average of moisture, total ash, acid insoluble ash, and water-soluble leachate in the 12 batches were 10.5 %, 13.8 %, 0.65 %, and 30.2 %, respectively;The average polysaccharide content is 4.3 %. CONCLUSIONS: The test standards can be used for quality control of Inonotus obliquus(Fr.) Pilat .
