摘要
分析和比较了曼氏无针乌贼墨汁黑色素(MelaninfromSepiellaMaindroniInk,MSMI)和虎斑乌贼墨黑色素(MelaninfromSepiapharaonisInk,MSPI)对D-半乳糖糖基化模型小鼠心脏内糖基化产物的影响.将80只ICR小鼠随机分为8组,包括空白对照组、衰老模型组以及2种乌贼墨汁黑色素低剂量组(120(mg·kg^-1)·d^-1)、中剂量组(240(mg·kg^-1)·d^-1)、高剂量组(480(mg·kg^-1)·d^-1).空白对照组皮下注射生理盐水,其他组皮下注射200(mg·kg^-1)·d^-1的半乳糖,连续灌胃45d后,对小鼠心脏组织中AGEs、RAGE、esRAGE、sRAGE及血清中Hb-AGE的含量进行检测.结果表明,2种乌贼墨汁黑色素具有清除AGEs及其受体、血清中的Hb-AGE的能力,作用效果与摄入的剂量成正比;其中低剂量与中剂量的MSMI清除血清中的Hb-AGE的效果更明显,但相同高剂量的MSPI清除血清中Hb-AGE的效果更明显.总体看MSMI对心脏组织中的AGEs的清除效果更强、对机体合成RAGE、sRAGE、esRAGE的抑制更为明显.
The current study was designed to compare the scavenging effects of two melanins, melanin from Sepiella maindroni ink(MSMI) and melanin from Sepia pharaonis ink(MSPI), on glycosylation end products and receptors. ICR mice were the subjects and randomly divided into eight groups, including control group, D-galactose group, MSMI treated groups with low(120(mg·kg^-1)·d^-1), medium(240(mg·kg^-1)·d^-1) and high(480(mg·kg^-1)·d^-1) dosage, MSPI treated groups with low(120(mg·kg^-1)·d^-1), medium(240(mg·kg^-1)·d^-1) and high(480(mg·kg^-1)·d^-1) dosage. Except for the control group, the remaining groups were treated with 200(mg·kg^-1)·d^-1 D-galactose with subcutaneous injection. A series of indicators were evaluated to determine the effects of melanin as AGEs, RAGE, esRAGE, sRAGE and serum Hb-AGE. The results showed that the two types of squid ink melanin had the ability to clear AGEs and its receptors, and Hb-AGE in serum. The effect was dose dependent. The effect of low-and medium-dose MSMI in clearing serum Hb-AGE was more pronounced, but the high dose of MSPI showed the most significant effects. Overall, MSMI has a greater effect on the clearance of AGEs in heart tissue and inhibition of RAGE, sRAGE and esRAGE to a greater extent. These results indicate that MSMI has an effective anti-aging effect.
