血府逐瘀汤对冠心病模型大鼠血管及心肌细胞凋亡的影响 被引量:10

Effects of Xuefu Zhuyu Decoction on Apoptosis of Blood Vessels and Myocardial Cells in Coronary Heart Disease Model Rats

摘要 目的:观察血府逐瘀汤对冠心病模型大鼠的血管重塑效应及对心肌细胞凋亡的影响。方法:将30只SD大鼠随机分为空白组、模型组及干预组,每组10只。模型组及干预组大鼠接受高脂饲料喂养7周以制备冠心病模型,造模后中药干预组大鼠接受血府逐瘀汤灌胃,模型组及空白组大鼠接受等剂量生理盐水灌胃,连续灌胃4周,干预结束后测量3组大鼠心功能及冠状动脉血流量,随后将大鼠处死后取冠状动脉及左心室组织,进行HE染色观察冠状动脉及左心室组织的病理学变化。采用Westernblotting法测定PI3K、AKT、Bcl-2、Bax的表达差异。结果:1)与空白组比较,模型组LVSP、-dp/dtmax、+dp/dtmax绝对值明显下降,差异有统计学意义(P<0.05),其中干预组上述指标较模型组高;2)与假手术大鼠心肌组织比较,模型组及干预组大鼠左心室心肌均出现明显的病理损害征象;与模型组大鼠比较,干预组心肌组织受损程度明显减轻;与空白组比较,模型组及干预组大鼠冠状动脉内中膜均出现明显增厚,经过血府逐瘀汤干预后的干预组大鼠动脉内中膜厚度比值明显低于模型组,差异有统计学意义(P<0.05);3)Westernblotting法检测结果显示:与空白组比较,2组接受造模的大鼠心肌组织PI3K、Bcl-2表达降低,Bax升高,血府逐瘀汤可上调PI3K及AKT的磷酸化程度,提高了Bcl-2表达,抑制了Bax的水平。结论:血府逐瘀汤有抑制冠心病模型大鼠血管重塑及保护心脏的效应,其作用机制可能与介导PI3K-AKT信号通路有关。 Objective: To observe the effects of Xuefu Zhuyu Decoction on vascular remodeling and cardiomyocyte apoptosis in coronary heart disease model rats. Methods: A total of 30 SD rats were randomly divided into sham operation group,model group and intervention group,with 10 rats in each group.Rats in the model group and intervention group were fed with high-fat diet for 7 weeks to prepare coronary heart disease model.Rats in the intervention group received Xuefu Zhuyu Decoction by gavage.Rats in the model group and the sham operation group received the same dose of normal saline by gavage for 4 weeks.After the intervention,cardiac function and coronary blood flow were measured in each group of rats,and the coronary artery and left ventricular tissue were taken after the rats were killed.Pathological changes of the coronary artery and left ventricular tissue were observed by HE staining.Expressions of PI3K,AKT,Bcl-2 and Bax were detected by Western blotting. Results: 1)Compared with the sham operation group,the absolute values of LVSP,-dp/dtmax and+dp/dtmax in the model group were decreased obviously,with statistical significance( P <0.05),in which the above indexes in the intervention group were up-regulated compared with those in the model group. 2)The left ventricular myocardium in the model group and the intervention group showed obvious pathological changes compared with that in the sham operation group.Compared with the model group,the damage degree of myocardial tissue in the intervention group was significantly reduced;compared with the sham operation group,the intima-media thickness of coronary artery in the model group and the intervention group was significantly thickened.After the intervention of Xuefu Zhuyu Decoction,the ratio of intima-media thickness of the artery in the intervention group was significantly lower than that in the model group,and the difference was statistically significant( P <0.05). 3)Western blotting showed:compared with the sham operation group,the expressions of PI3K and Bcl-2 in the myocardial tissue of rats in the other 2 groups underwent modeling were decreased,while the expression of Bax was increased.Xuefu Zhuyu Decoction could up-regulate the phosphorylation of PI3K and AKT,up-regulate the expression of Bcl-2,and inhibit the level of Bax. Conclusion: Xuefu Zhuyu Decoction has the effect of inhibiting vascular remodeling and protecting the heart in coronary heart disease model rats,and its mechanism may be related to mediating the PI3K-AKT signaling pathway.
