中医烙法对慢性扁桃体炎扁桃体组织中细胞因子表达影响的实验研究

Experimental study on the effect of TCM Cauterization on the expression of cytokines in tonsil tissue of chronic tonsillitis

目的观察中医烙法对慢性扁桃体炎患者扁桃体组织中细胞因子表达的干预效果,探讨中医烙法对慢性扁桃体炎患者抗炎作用机制。方法在临床接受中医烙法治疗的慢性扁桃体炎患者中,采用随机数字法随机抽取10名患者,在中医烙法首次治疗前及末次治疗次后分别钳取扁桃体组织作为烙治组(烙前组、烙后组),并选取10名特发性扁桃体肥大正常人扁桃体组织作为健康对照组。采用酶联免疫吸附(ELISA)法检测中医烙法治疗前后及正常人扁桃体组织中IL-4、IL-5、IL-10、IFN-γ、TGF-β含量。结果各组扁桃体组织中IL-4、IL-5、IL-10、IFN-γ、TGF-β含量检测结果显示:与对照组比较,烙治组(烙前组、烙后组)扁桃体组织中IL-4、IL-5、IL-10、IFN-γ、TGF-β含量均显著升高,有统计学差异(P<0.01);试验组中,与治疗前比较,治疗后的扁桃体组织中IL-4、IL-5、IL-10、IFN-γ、TGF-β含量均不同程度降低,差异有统计学意义(P<0.01)。结论中医烙法对慢性扁桃体炎具有抗炎作用,其作用机制可能是通过抑制扁桃体组织中IL-4、IL-5、IL-10、IFN-γ、TGF-β等细胞因子表达水平实现的。

Objective To observe the interventional effect of TCM Cauterization method on the expression of cytokines in tonsillar tissue of patients with chronic tonsillitis .To discuss the mechanism of anti- inflammation in patients with chronic tonsillitis by TCM Cauterization method. Methods Among patients with chronic tonsillitis treated by TCM Cauterization method, a randomized digital method was used to select 10 patients randomly . Tonsil tissue was removed before the first treatment and after the last treatment of TCM cauterization method as cautery group (pre-cautery group, post-cautery group). Ten normal people with idiopathic tonsillitis were selected as healthy control group. The contents of IL-4, IL-5, IL-10, IFN-γ and TGF-β in tonsil tissues of normal people before and after treatment by traditional Chinese medicine were detected by ELASA. Results The contents of IL-4, IL-5, IL-10, IFN-γ and TGF-β in tonsil tissues of each group were detected. Compared with the control group, the contents of IL-4, IL-5, IL-10, IFN-γ and TGF-β in tonsil tissues of the cautery group (pre-cautery group and post-cautery group) were all significantly increased and difference was statistically significant(P<0.01). In the experimental group, the levels of IL-4, IL-5, IL-10, IFN-γ and TGF-β in tonsil tissues after treatment were reduced to different degrees compared with those before treatment, the difference was statistically significant (P<0.01). Conclusion TCM Cauterization method has anti-inflammatory effect on chronic tonsillitis, and its mechanism may be realized by inhibiting the expression levels of cytokines such asIL-4, IL-5, IL-10, IFN-γ and TGF-β in tonsil tissues.
