摘要
食源性致病菌是导致肉类食品安全突发事件的主要因素,因此采用一定检测手段来控制食源性致病菌非常有必要。传统的细菌分离、培养与鉴定耗时较长,特别是有的细菌难以培养,因而快速、简便、特异的检测方法成为食源性致病菌检测的热点。本文通过SELEX筛选出一系列关于沙门氏菌、单增李斯特菌、金黄色葡萄球菌的ssDNA适配子,这些适配子与目的菌结合,建立了该检测方法的分子识别基础。将筛选出来的核酸序列经巯基修饰后再与本研究所合成的GNPs孵育结合,以GNPs在不同状态下颜色的变换为表现方式,建立了3种常见肉制品中致病菌金纳米适配子检测方法,结果肉眼可见,不需要借助任何仪器,具有检测准确、操作简便、耗时较短等优点。
Food-borne pathogens are the main cause of safety incidents in meat products,so it is necessary to use certain detection methods to control food - borne pathogens. Traditional bacteria isolation, culture and identification take a long time,especially some bacteria are difficult to culture,so the rapid,simple and specific detection method has become a hot spot for food -borne pathogen detection. In this paper, a series of ssDNA aptamers for Salmonella, Listeria monocytogenes and Staphylococcus aureus were screened by SELEX. These aptamers were combined with the target bacteria to establish the molecular recognition basis of the detection method. The selected nucleic acid sequence was incubated with thiol and then incubated with the GNPs synthesized in this study. The color of GNPs was transformed into different expressions to establish the pathogens of three common meat products. This detection method is visible to the naked eye,and does not require any instrument,and has the advantages of accurate detection,simple operation,and short time consumption.
