低强度激光治疗改善小鼠脑梗死后认知障碍的机制研究 被引量:2

摘要 目的探讨低强度激光治疗对脑梗死后认知障碍小鼠的影响及可能的作用机制。方法采用线栓法制作小鼠脑梗死后认知障碍模型,采用随机数字表法将实验小鼠分为假手术组(Sham组)、脑梗死后认知障碍组(PSCI组)和脑梗死后认知障碍低强度激光治疗组(PSCI+LLLI组);采用短暂性大脑中动脉阻塞建立小鼠的脑梗死后认知障碍模型,各组小鼠在相同的环境下分别饲养28 d后采用水迷宫实验检测小鼠认知功能,采用ELISA法检测小鼠前额叶皮层的ATP和线粒体复合物含量。结果与Sham组小鼠比较,PSCI组小鼠水迷宫成绩显著下降(P<0.05),PSCI组小鼠前额叶皮层ATP含量和线粒体复合物的含量降低;PSCI+LLLI组小鼠水迷宫成绩较PSCI组有显著改善(P<0.05);与PSCI组小鼠比较,PSCI+LLLI组小鼠脑内ATP和线粒体复合物的含量显著增加。结论低强度激光治疗可以改善脑梗死后认知障碍小鼠的认知功能,其作用机制可能与提高小鼠脑内ATP含量有关。 Objective To observe and explore the effects and the possible mechanisms of low-level laser irradiation on post-stroke cognitive impairment(PSCI)in mice.Methods Mice were divided into 3 groups: sham-operated(Sham)group,PSCI group and PSCI+Low-level laser irradiation(PSCI+LLLI)group.Middle cerebral artery occlusion(MCAO)was used to establish cognitive impairment model.After breeded under the same environment for 28 days,water maze experiment was used for the evaluation of cognitive impairment. The level of ATP and mitochondrial complex in prefrontal cortex(PFC)was detected by Elisa Kit.Results Compared with the Sham group,the PSCI group showed significantly poorer water maze performance.The level of ATP and mitochondrial complex decreased significantly in PSCI group.When compared with PSCI group,LLLI could improved the cognitive impairment in water maze experiment,and the concentrations of ATP and mitochondrial complex were improved significantly.Conclusion By improving the concentration of ATP in PFC,LLLI could improve the cognitive function of PSCI in mice.
