摘要
目的探讨睾酮对结核分枝杆菌(Mycobacterium tuberculosis,MTB)感染的鼠源巨噬细胞RAW264.7细胞自噬的影响及其分子机制。方法MTB感染RAW264.7细胞24h后,给予10-8mol/L睾酮处理24h;采用透射电镜观察细胞自噬情况,Westernblot法检测细胞自噬相关蛋白以及MAPKs信号通路蛋白。结果睾酮干预MTB感染的RAW264.7细胞后自噬体、自噬特异标志物LC3Ⅱ均明显增加(P<0.01),通路蛋白JNK磷酸化水平明显上调(P<0.001)。结论睾酮可能通过激活JNK信号通路诱导MTB感染的RAW264.7细胞自噬。
To investigate the effect of testosterone on the autophagy of RAW264.7 cells infected with Mycobacterium tuberculosis (MTB) and its molecular mechanism,RAW264.7 cells were infected with MTB for 24 h, and then treated with 10-8 mol / L testosterone for 24 h.The autophagy was observed by transmission electron microscopy;the autophagy-related proteins and MAPKs signal pathway proteins were detected by Western blotting. Data showed that testosterone intervention significantly increased the autophagosome and autophagy specific marker LC3Ⅱ(P<0.01),and significantly up-regulated the pathway protein JNK phosphorylation level (P<0.001) in MTBinfected RAW264.7 cells.In conclusion,testosterone may induce autophagy in MTB-infected RAW264.7 cells by activating JNK signaling pathway.
