子宫内膜癌患者组织中lncRNA PVT1、H19和TUG1表达水平及临床意义 被引量:4

Expression Levels and Clinical Significances of Long Non-Coding RNA PVT1,H19 and TUG1 in Tissues of Endometrial Carcinoma Patients

摘要 目的探讨子宫内膜癌组织中长链非编码RNA PVT1、H19和TUG1的表达水平,并评估其对子宫内膜癌患者的预后价值。方法收集四川省人民医院2013年12月-2015年5月期间实施手术治疗的54例子宫内膜癌组织标本及癌旁组织标本,另外选取46例因其他疾病切除子宫的正常子宫内膜组织标本作为正常组;分别使用Real-time PCR检测子宫内膜癌组织、癌旁组织以及正常子宫内膜组织中PVT1、H19和TUG1的表达水平,并统计分析各组患者临床病理特征。采用Kaplan-Meier对患者生存情况进行分析,Cox回归模型分析PVT1、H19和TUG1表达、患者年龄、病理分期、分化程度、肌层浸润情况以及是否出现淋巴结转移与子宫内膜癌患者预后关系。结果与正常组和癌旁组相比,子宫内膜癌组中PVT1、H19和TUG1的表达水平均明显升高(P<0.05)。子宫内膜癌组中PVT1、H19和TUG1表达均与年龄无关系(P>0.05);PVT1表达高低与病理分期、分化程度、肌层浸润情况以及是否出现淋巴结转移有关(P<0.05);H19和TUG1表达高低与肌层浸润情况和是否出现淋巴结转移有关(P<0.05)。PVT1、H19和TUG1高表达者预后存活期明显低于低表达者(P<0.05)。PVT1、H19和TUG1表达、病理分期、分化程度、肌层浸润情况以及淋巴结转移子是影响子宫内膜癌预后的独立因素。结论长链非编码RNA PVT1、H19和TUG1在子宫内膜癌患者中均呈现高表达,且与临床病理参数和预后密切相关,推测三者可能参与子宫内膜癌发生发展,可作为诊断及评估预后的标志物。 Objective To investigate the expression levels of long non-coding RNA PVT1, H19 and TUG1 in endometrial carcinoma tissues and to evaluate their prognostic value in endometrial carcinoma. Methods From December 2013 to May 2015 , 54 specimens of endometrial carcinoma tissues and para cancerous tissues were collected, and 46 normal endometrium specimens were selected as normal groups;the expression levels of PVT1 , H19 and TUG1 in endometrial carcinoma tissues, para cancerous tissues and normal endometrium tissues were detected by Real-time PCR, the clinicopathological features of the patients were analyzed statistically, Kaplan-Meier was used to analyze the survival of patients, the Cox regression model was used to analyze the relationships between the expressions of PVT1, H19 and TUG1, age, the pathological stage, differentiation, myometrial invasion and lymph node metastasis with the prognosis of patients with endometrial carcinoma. Results Compared with the normal group and the paracancerous group, the expression levels of PVT1, H19 and TUG1 in endometrial carcinoma group were significantly increased ( P<0. 05 ). The expressions of PVT1, H19 and TUG1 in endometrial carcinoma group were not related to age ( P>0. 05);the expression of PVT1 was related to pathological stage, differentiation, myometrial invasion and lymph node metastasis ( P<0. 05 );the expression levels of H19 and TUG1 were related to myometrial invasion and lymph node metastasis ( P<0. 05 ). The survival time of patients with high expressions of PVT1 , H19 and TUG1 was significantly lower than those with low expressions ( P<0. 05 ). The expressions of PVT1, H19 and TUG1, pathological stage, differentiation, myometrial invasion and lymph node metastasis were independent prognostic factors of endometrial carcinoma. Conclusion Long non-coding RNA PVT1, H19 and TUG1 are highly expressed in endometrial carcinoma patients, and it is closely related to the clinicopathological parameters and prognosis, it is speculated that the three may participate in the occurrence and development of endometrial carcinoma, and can be used as a marker for diagnosis and prognosis.
