摘要
目的探讨姜黄素对人肝癌HepG2细胞增殖和侵袭转移的影响及其相关机制。方法应用10、20、40、60μmol/L姜黄素处理肝癌细胞株HepG2;采用MTT检测不同浓度的姜黄素对HepG2细胞增殖的影响;运用Transwell侵袭转移实验测定不同浓度的姜黄素对HepG2细胞体外侵袭能力的影响。采用Westernblot方法检测经不同浓度姜黄素处理后,HepG2细胞内细胞周期素D1(CyclinD1)、基质金属蛋白酶2(matrix metalloproteinase 2,MMP-2)和E-钙黏素(E-cadherin)蛋白表达。结果姜黄素能明显抑制肝癌HepG2细胞的增殖和体外侵袭能力,且具有显著剂量依赖性。肝癌HepG2细胞中CyclinD1、MMP-2蛋白表达随着姜黄素的浓度的增多而明显的降低,而E-cadherin蛋白表达则随着姜黄素剂量的增多而显著升高。结论姜黄素明显抑制肝癌HepG2细胞的增殖和侵袭转移能力,与下调CyclinD1、MMP-2和上调E-cadherin的蛋白表达相关。
Objective To investigate the effect of curcumin on the proliferation, invasion and metastasis of human hepatocellular carcinoma HepG2 cells and its related mechanism. Methods HepG2 cells were exposed to control(medium) or 10, 20, 40, 60μmol/L curcumin. The effect of curcumin on the proliferation of HepG2 cells at different concentrations were detected by MTT. The invasive abilities of HepG2 cells were detected by Transwell chamber. The expression of CyclinD1, MMP-2, E-cadherin proteins in HepG2 cells was detected by Western blot. Results Curcumin evidently inhibited the proliferation and invasion of HepG2 cells in vitro in a dose-dependent manner. The expression level of CyclinD1 and MMP-2 proteins in HepG2 cells decreased with the increase of curcumin concentration, while the expression level of E-cadherin protein increased with the increase of curcumin. Conclusion can significantly The inhibition of curcumin on the proliferation, invasion and metastasis of hepatocellular carcinoma HepG2 cells is related to down-regulation of CyclinD1, MMP-2 and up-regulation of E-cadherin expression.
