芹菜素对喉癌Hep-2细胞增殖、迁移与侵袭的影响

Effect and possible mechanism of apigenin on proliferation and apoptosis of laryngeal cancer Hep-2 cells

目的探讨芹菜素对喉癌Hep-2细胞增殖、迁移与侵袭的影响及可能机制。方法MTT法检测不同浓度的芹菜素(0、20、40、80μmol/L)对喉癌Hep-2细胞增殖的影响。40μmol/L芹菜素处理Hep-2细胞48h后,Transwell实验检测Hep-2细胞体外迁移和侵袭能力变化;实时PCR和Westernblot检测Hep-2细胞Wnt1、β-catenin、环氧合酶2(COX2)与基质金属蛋白酶2(MMP-2)的蛋白与mRNA的表达水平。结果芹菜素抑制喉癌Hep-2细胞增殖,并呈时间浓度依赖性;40μmol/L芹菜素处理Hep-2细胞48h能够有效抑制Hep-2细胞的迁移和侵袭能力,降低Wnt1、β-catenin、COX2与MMP-2mRNA相对表达量和蛋白表达水平。结论芹菜素抑制喉癌细胞的增殖、迁移和侵袭能力与抑制喉癌Hep-2细胞中Wnt/β-catenin信号通路相关。

Objective To investigate the effect of apigenin on proliferation, migration and invasion of laryngeal cancer Hep-2 cells and its possible mechanism. Methods MTT assay was used to detect the effects of apigenin(0, 20, 40, 80 μmol/L) on the proliferation of laryngeal cancer Hep-2 cells. 48 hours after treatment with 40 μmol/L apigenin, the migration and invasion ability of Hep-2 cells were detected by Transwell assay, and the expressions of Wnt1, beta-catenin, cyclooxygenase 2(COX2) and matrix metalloproteinase 2(MMP-2) were detected by real-time PCR and Western blot. Results Apigenin inhibited the proliferation of laryngeal cancer Hep-2 cells in a time-and concentration-dependent manner. Apigenin(40 micromol/L for 48 hours) effectively inhibited the migration and invasion of Hep-2 cells, reduced the levels of Wnt1, beta-catenin, COX2 and MMP-2. Conclusion The inhibition effect of Apigenin on the proliferation, migration and invasion of Hep-2 cellsis related to inhibiting Wnt/beta-catenin signaling pathway.
