芹菜素对肺癌PC9GR细胞侵袭和迁移的抑制作用及机制 被引量:1

摘要 目的探讨芹菜素对肺癌PC9GR细胞侵袭和迁移能力的影响,并探究可能的相关分子机制。方法将对数生长期细胞随机分组,实验组分别加入含有10μmol/L、20μmol/L芹菜素的培养基,阴性对照组加入含0.1%DMSO的完全培养基。培养48h后,CCK-8法检测细胞存活率,Transwell实验检测细胞侵袭和迁移能力,Westernblot检测E-cadherin等相关蛋白的表达。结果与阴性对照组比较,10μmol/L组和20μmol/L组细胞存活率有所降低,细胞侵袭和迁移明显减少(均P<0.05)。实验组中E-cadherin蛋白表达显著上调,Snail、MMP-9、VEGF的表达明显减少(均P<0.05),对N-cadherin、Slug及MMP-2的表达影响不明显。结论芹菜素可明显降低肺癌PC9GR细胞的侵袭和迁移能力,并且可能与上调细胞中E-cadherin的表达,减少Snail、MMP-9及VEGF表达有关。 Objective To investigate the effects and molecular mechanisms of apigenin on invasion and migration of PC9GR lung cancer cells.Methods Cells were randomly divided into groups.The experimental group was added with medium containing 10μmol/L or 20μmol/L of apigenin,and the negative control group was medium containing 0.1%DMSO.Cell viability was detected by CCK-8 method 48 hours later.Transwell migration assay was used to detect the ability of cell invasion and migration.Western blotting was used to detect the expression of E-cadherin and other related proteins.Results Compared with control group,cell viabilities in the 10μmol/L and the 20μmol/L groups were slightly reduced.The cell invasion and migration in these groups were both significantly decreased(all P<0.05).The expression of E-cadherin was markedly enhanced in the experimental group,while the expressions of Snail,MMP-9 and VEGF were weakened(all P<0.05).Expressions of N-cadherin,Slug and MMP-2 were not affected obviously in these groups.Conclusion Apigenin can significantly reduce the invasion and migration of PC9GR lung cancer cells,which may be related to up-regulation of E-cadherin and down-regulation of Snail,MMP-9 and VEGF.
