Expression of VGLL4 in colon cancer and its significance

目的分析VGLL4在结肠癌组织内的表达情况,探讨VGLL4在不同临床病理组织学结肠癌患者中的表达差异及其与预后的关系,以期为结肠癌的临床诊疗提供参考依据。方法收集120例结肠癌根治术患者标本,采用免疫组化法比较癌组织及癌旁组织的VGLL4表达情况,计算VGLL4的光密度值(IOD)值中位数,并依据其将患者分为VGLL4高表达组及低表达组。分析VGLL4的表达水平并探讨其与结肠癌患者临床、病理组织学及预后的关系。结果采用免疫组化法显示VGLL4在癌组织中的表达水平显著低于癌旁组织,差异有统计学意义(P<0.05)。对VGLL4在不同病理组织学结肠癌患者中的表达差异分析显示,VGLL4高表达与低表达组患者在性别、年龄、BMI、M分期上比较无统计学差异(P>0.05);而在肿瘤直径、T分期及N分期上比较差异有统计学意义(P<0.05)。采用Logistic回归模型对VGLL4表达的独立影响因素进行分析得出,T分期、肿瘤直径及TNM分期是VGLL4高表达的独立影响因素。VGLL4高表达组患者血清CEA水平显著低于低表达组,差异有统计学意义(P<0.05)。VGLL4高表达组患者血清中CA199水平与低表达组比较,差异无统计学意义(P>0.05)。Spearman相关性分析显示,结肠癌患者VGLL4表达水平与血清CEA水平呈显著负相关(r=-0.607,P<0.01)。VGLL4低表达组5年累积复发率高于高表达组,而5年累积死亡率低于高表达组(P<0.05);结论VGLL4在结肠癌组织表达水平显著低于癌旁组织,与肿瘤进展程度相关,且其水平降低提示预后不良。

Objective To analyze the expression of VGLL4 in colon cancer tissues,and to investigate the differential expression of VGLL4 in different clinicopathological histological cancer patients and its relationship with prognosis in order to provide reference for clinical diagnosis and treatment of rectal cancer.Methods The clinical data of 120 patients who underwent radical resection of rectal cancer in our hospital from January 2010 to January 2013 were included.Immunohistochemistry was used to compare the expression of VGLL4 in cancer tissues and adjacent tissues, and calculate the expression of VGLL4.The median value of optical density ( IOD) was divided into VGLL4 high expression group and low expression group.The expression level of VGLL4 was analyzed and its relationship with clinical,histopathology and prognosis of rectal cancer patients was discussed.Results Immunohistochemistry showed that the expression level of VGLL4 in cancer tissues was significantly lower than that in adjacent tissues ( P< 0.05).The difference of expression of VGLL4 was observed in different histopathological rectal cancer patients.The analysis showed that there was no significant difference in gender,age,BMI and M staging between the high-and lowexpression groups of VGLL4 ( P>0.05),but the difference in tumor diameter,T stage and N stage was statistically significant ( P<0.05);Logistic regression model was used to analyze the independent influencing factors of VGLL4 expression.T stage,tumor diameter and TNM stage were independent factors affecting the high expression of VGLL4. Serum CEA levels were significantly lower in patients with high expression of VGLL4.than that of the lower expression group ( P< 0.05 ).There was no significant difference in serum CA199 level between the VGLL4 high expression group and the low expression group ( P> 0.05).Spearman correlation analysis showed that in rectal cancer patients there was a significant negative correlation between VGLL4 expression level and serum CEA level ( r =- 0.607,P< 0.01).The 5-year cumulative recurrence rate of VGLL4 low expression group was higher than that of high expression group,and the 5-year cumulative survival rate was lower than that of high expression group ( P < 0.05 ). Conclusions VGLL4 expression level in rectal cancer tissues is significantly lower than that of adjacent tissues,and related to tumor progression.The decrease in its level suggests a poor prognosis.
