摘要
目的探讨微小RNA-216b(microRNA-216b,miR-216b)在急性髓系白血病(acute myelogenous leukemia,AML)中的表达及意义。方法急性髓细胞白血病患者53例作为AML组,同期31例原发性免疫性血小板减少症(Primary immune thrombocytopenia,ITP)患者作为对照组。提取骨髓单个核细胞,检测miR-216b的表达水平,分析其与临床特征的关系。结果 AML组miR-216b相对中位表达水平高于对照组,差异有统计学意义( P <0.05);SML患者不同FAB分型、染色体核型和基因突变类型miR-216b表达水平比较差异无统计学意义( P >0.05);根据miR-216b相对中位表达水平,以0.345作为分界,将53例 AML患者分为高表达和低表达组;高表达组和低表达组在性别、年龄、血红蛋白、血小板计数和完全缓解率差异无统计学意义( P >0.05);高表达组WBC计数、白细胞计数和骨髓原始细胞比例低于低表达组,1年内完全缓解后复发率和1年死亡率均高于低表达组,差异均有统计学意义( P <0.05)。结论miR-216b在AML患者中表达水平高于对照组,与疾病复发和生存有一定的关联。
Objective To investigate the expression and significance of microRNA-216 b(miR-216 b) in acute myelogenous leukemia(AML).Methods A total of 53 patients with AML were enrolled as AML group,at the same time, the other 31 patients with primary immune thrombocytopenia(ITP) were enrolled as control group,the patients in both groups were treated in our hospital from January 2014 to June 2017.The expression levels of miR-216 b in the extracted bone marrow mononuclear cells were detected,and its correlation with clinical features was analyzed.Results The relative median expression levels of microRNA-216 b in AML group were significantly was higher than those in control group(P<0.05). There were no significant differences in the expression levels of microRNAs-216 b in AML patients with different FAB typing,chromosomal karyotype and gene mutation type(P>0.05).Taking 0.345 as the boundary of microRNAs-216 b expression,the patients in AML group were redivided into high expression group and low expression group. There were no significant differences in the gender,age,hemoglobin,platelet count and complete remission rate between high expression group and low expression group(P>0.05).However the WBC count,white blood cell count and the proportion of bone marrow primordial cells in high expression group were significantly lower than those in low expression group,but the recurrence rate and 1-year mortality after complete remission within one year in high expression group were significantly higher than those in low expression group(P<0.05).Conclusion The expression levels of miR-216 b in patients with AML are higher than those of patients with ITP,which is associated with disease recurrence and survival.
