过氧乙酸对大鼠血清炎性因子水平及肺组织结构的影响

Effect of Peroxyacetic Acid on the Levels of Serum Inflammatory Factors and the Structure of Lung Tissue in Rats

目的观察过氧乙酸(PAA)喷雾消毒对大鼠血清肿瘤坏死因子-α(TNF-α)、白细胞介素6(IL-6)含量及肺组织结构的影响。方法将72只SD大鼠随机分为对照组(A组,0. 9%氯化钠溶液)和PAA低、高剂量(0. 2%、0. 5%)组(B_1、B_2组),每组24只;再分为1周,2周,4周3个亚组,每组8只。将大鼠置离地面1 m处的密闭空间中,按8 mL/m^3剂量以药物相应体积分数进行喷雾,1 min喷完,30 min后将大鼠转至屏障动物房中,每日1次,连续2周。然后停药2周,第4周时结束实验。以酶联免疫吸附法测定大鼠血清TNF-α和IL-6含量,观察大鼠肺组织结构。结果与A组比较,B_1组和B_2组雄性和雌性大鼠给药2周、停药2周内体质量和血清TNF-α含量均无显著差异(P> 0. 05),B_1组和B_2组大鼠给药1周时血清IL-6含量均显著增加(P <0. 05);A组大鼠肺组织结构清晰,B_1组和B_2组大鼠肺组织结构紊乱,与A组比较差异显著。结论 PAA喷雾消毒对实验大鼠血清中IL-6含量及肺组织结构有明显影响,可作为评价PAA对呼吸系统损伤的相关参考数据。

Objective To observe the effect of spray disinfection with peroxyacetic acid ( PAA ) on the levels of serum tumor necrosis factor -α( TNF -α),interleukin -6 ( IL -6 ) and the structure of lung tissue in rats. Methods Totally 72 SD rats were randomly divided into the control group ( group A,0. 9% sodium chloride solution ) and PAA low dosage ( 0. 2%),high dosage ( 0. 5%) groups ( group B 1 , B 2 ),24 rats in each group,and then they were divided into three subgroups: one - week subgroup,two - week subgroup and four - week subgroup,8 rats in each group. The rats were placed in the confined space at 1m of the ground and sprayed with the corresponding volume fraction of drugs according to the dosage of 8 mL /m 3 in 1 min. After 30 min,the rats were transferred to the barrier environ- ment,and the drug was sprayed once a day for two weeks. Then the drug was stopped spraying for two weeks and the experiment was completed at the 4th week. The levels of TNF -α and IL - 6 in the serum of rats were measured by enzyme - linked immunosorbent assay ( ELISA ),and the structure of lung tissue of rats was observed. Results Compared with those in group A,the body weight and the levels of serum TNF -α of the male and female rats in group B 1 and group B 2 after 2 weeks of administration and 2 weeks within withdrawal had no significant difference ( P > 0. 05 ). Compared with those in group A,the levels of serum IL - 6 of rats in group B 1 and group B 2 increased significantly after 1 week of administration ( P < 0. 05 ). The lung tissue structure of rats in group A was clear, and that in group B 1 and group B 2 was disordered,that in group B 1 and group B 2 was significantly different from that in group A. Conclusion Spray disinfection with PAA has significant effect on the levels of serum IL - 6 and the structure of lung tissue in experi- mental rats,which can be used as reference data for the evaluation of PAA on respiratory system injury.
