摘要
以柳蒿芽为原料,在单因素试验的基础上通过Plackett-Burman试验和Box-Behnken试验确定最优提取方案,并利用D101大孔吸附树脂进行纯化,最终得到柳蒿芽多酚。结果表明:柳蒿芽多酚的最佳提取工艺为乙醇浓度70%、酶解温度60℃、酶解pH 5.5、加酶量50 mg/g、酶解时间2 h、料液比1∶30(g/mL)、超声时间30 min、超声功率400 W,在此条件下,柳蒿芽多酚得率为9.41%,纯化后的柳蒿芽多酚纯度为76.2%。体外抗氧化试验结果表明,柳蒿芽多酚质量浓度为1.2 mg/mL时,Fe3+还原力和DPPH+清除力最强,分别为1.09和94.20%,且高于维生素C对照组,说明柳蒿芽多酚抗氧化活性较强。
With Artemisia integrifolia L.as raw material,based on the single factor experiment,the optimum extraction scheme was determined by Plackett-Burman test and Box-Behnken test,and puri fied by D101 macroporous adsorption resin to obtain the polyphenols from Artemisia integrifolia L..The results showed that the optimum extraction conditions were as follows:the concentration of ethanol 70%,enzymolysis temperature 60℃,enzymolysis pH 5.5,amount of enzyme 50 mg/g,enzymolysis time 2 h,ratio of material to liquid 1∶30(g/mL),ultrasonic time 30 min,ultrasonic power 400W.The yield of polyphenol was 9.41%,and the purity of polyphenols was 76.2%.The results of invitro antioxidant test showed that AILP had the strongest Fe 3+reducing power and DPPH+scavenging ability of 1.09 and 94.20%,when it was 1.2 mg/mL,higher than Vitamin at the same concentration levels.Respectively,the antioxidant activity of AILP was very strong.
